摘 要:目的探讨溪黄草有效成分nodosin体外保留灌注的方式对移植肝病理损伤的影响。方法建立nodosin体外保留灌注大鼠肝脏的实验模型。实验组分为nodosin高剂量组(200μg/ml)、中剂量组(20μg/ml)、低剂量组(10μg/ml)和空白对照组(乳酸林格注射液)进行体外灌注。采用Banff标准分级评价体外保留灌注的离体大鼠肝脏的病理损伤。电镜下评估nodosin高剂量组肝脏细微结构的损伤情况。建立使用nodosin(200μg/ml)保留灌注供肝行大鼠原位肝移植的实验模型,设立空白对照组,分别于移植术后1d、3d、7d、21d获取肝脏病理组织,采用Suzuki评分标准评价移植肝的病理损伤。结果 nodosin各剂量组Banff评分与空白对照组比较差异无统计学意义。电镜下,no-dosin高剂量组保留灌注2h的大鼠肝脏组织结构无明显破坏。移植模型中,移植术后1d、3d、7d两组的Suzuki评分比较差异无统计学意义,但术后21d两组的Suzuki评分比较差异有统计学意义(P<0.01)。结论使用200μg/ml剂量以下的nodosin体外直接灌注不会对肝脏组织结构和肝细胞造成直接的病理损伤,是安全的。Objective To investigate the effect of nodosin(active constituent from Herba Isodi Lophanthoidis)perfusion on rat liver grafts pathological injury ex vivo.Methods The rat models reserved perfusion with nodosin were established ex vitro.Rat livers were randomly divided into four groups and underwent the following treatments:perfusion with nodosin-H(200 μg/ml),nodosin-M(20 μg/ml),nodosin-L(10 μg/ml)and lactic Ringer's(control)respecitively.Histological injury was measured by Banff criteria and the microstructure was observed using an electron microscope.Rat orthotopic liver transplantation(OLT)was performed using the rat livers perfused with lactic Ringer's(control)or nodosin(200 μg/ml)ex vivo.The histological injury was measured with Suzuki criteria on day 1,day 3,day 7 and day 21 after OLT.Results There was no significant difference in Banff scores between nodosin treatment group and control group,respectively.The grafts perfused with nodosin showed no obvious morphological damage under electron microscopy examination.On day 1,day 3,day 7 after OLT,there was no significant difference in Suzuki scores between nodosin(200 μg/ml)group and control group.On day 21 after OLT,there was statistical difference between the two groups(P0.01).Conclusion No histological damage is observed in the liver grafts perfused with nodosin(200 μg/ml)ex vivo.
