摘  要:研究了雷帕霉素发酵的最佳种子培养时间,发现60 h的种子液能够产生最高的发酵水平。系统地考察了发酵培养基中不同水平的碳氮源和各种无机盐对雷帕霉素发酵水平的影响,并确定最优的培养基配方。在以上优化后的条件下雷帕霉素原始菌的发酵水平从未优化时95 mg/L提高到170 mg/L左右,提高幅度达到79%,取得了明显的效果。然后通过多种诱变手段,经过多轮诱变育种,筛选到最高产量为420 mg/L的一株菌株,比出发菌株提高了3倍多。The optimal cultivation time of seed for rapamycin fermentation was investigated.It shows that the highest yield of fermentation with seed liquor can be achieved at 60 h.The effects of different medium components on rapamycin production were examined systematically,and one optimal fermentation medium was formulated.Under these optimal fermentation conditions,the yield of rapamycin is improved from 95 mg/L to 170 mg/L,achieving a big improvement by 79% with the above optimization.The systematic research of rational screening techniques finds an applicable way to generate high-yield strains from original microorganisms.The highest yield reaches 420 mg/L,about 3 times higher than that of original strain.
