摘　　要：目的 检测Bmi-1在胰腺癌组织中的表达,分析其与胰腺癌临床病理特征之间的关系.方法 实时荧光定量PCR法检测22例手术切除的新鲜胰腺癌组织及配对癌旁组织中Bmi-1 mRNA的表达;免疫组织化学法检测61例胰腺癌石蜡块标本中Bmi-1、p16蛋白的表达,并分析与临床病理指标的关系.结果 22例胰腺癌组织中Bmi-1 mRNA的相对表达量为0.314±0.040,与配对的癌旁组织0.143±0.056的表达量相差显著（P＜0.01）.61例胰腺癌组织中36例（59.0%）Rmi-1蛋白表达阳性,34例（55.7%）p16蛋白表达缺失.胰腺癌组织Bmi-1的阳性表达与淋巴结转移相关（P=0.025）;p16的表达缺失与淋巴结的转移（P=0.020）及肿瘤分化程度（P=0.018）相关;Bmi-1蛋白表达与p16蛋白表达呈负相关（P=0.033）.结论 Bmi-1在胰腺癌组织中表达增加,它可能通过抑制p16表达调控胰腺癌的恶性生物学行为.Objective To investigate the expression and significance of Bmi-1 in pancreatic cancer.Methods The expression of Bmi-1 mRNA was detected in pancreatic cancer and matched adjacent normal tissues by using real-time PCR in 22 cases of human pancreatic carcinoma.Bmi-1 and p16 protein were determined in 61 cases of human pancreatic carcinoma with immunohistochemistry and their relationship with clinicopathologic parameters was analyzed.Results The expression of Bmi-1 mRNA was 0.314 ±0.040 in cancerous tissues, and 0.143 ±0.056 in adjacent normal tissues, and the difference was statistically significant （P 〈0.01 ）.36 cases （59.0%） of PC were detected with positive expression of Bmi-1 protein, and 34 cases （55.7%） with no expression of p16 protein.The expression of Bmi-1 was significantly correlated with lymph node metastasis （ P = 0.020 ） and tumor differentiation （ P = 0.018 ）; the expression of Bmi-1 was negatively correlated with the expression of p16 （ P = 0.033 ）.Conclusions Expression of Bmi-1 was increased in pancreatic cancer, which may be involved in the malignant behavior of pancreatic cancer by inhibiting p16 expression.
