摘  要:目的探讨葡萄籽提取物原花青素(PA)对人肝癌细胞SMMC-7721细胞的诱导分化作用。方法用不同剂量的PA与SMMC27721细胞共同培养,以MTT法检测细胞增殖和PA对细胞增殖的抑制作用;镜下观察细胞形态改变;采用RT-PCR方法检测细胞甲胎蛋白mRNA表达,金氏法检测细胞碱性磷酸酶活性的变化。结果 PA在20~60μg/ml的浓度范围均能抑制细胞增殖,且抑制率随浓度的增加而增高(P<0.05),细胞形态和结构向正常肝细胞方向逆转,40μg/ml和60μg/ml的PA均可使细胞甲胎蛋白mRNA表达量明显下降,碱性磷酸酶活性明显升高。结论 PA对人肝癌SMMC27721细胞具有抗增殖和促分化作用。Objective To study the effect of grape seed procyanidin extract(PA) on the induction of differentiation of human hepatoma cell line SMMC-7721. Methods SMMC-7721 cells were cultured in RPM I-1640 and treated by PA at different doses. The proliferation of the cells was examined by MTT assay. Cell morphology was observed by light microscopy. The reverse transcriptase-polymerase chain reaction(RT-PCR) was used to detect the change of alpha fetoprotein(α-FP) mRNA. The specific activities of alkaline phosphatase(ALP) were assayed by ALP kit based on King's method. Results The cell proliferation was inhibited by PA at 20-60μg/ml in dose dependent manners. 40μg/ml PA induced the morphological normality of SMMC-7721 cells. After treated with 40μg/ml or 60μg/ml PA, the mRNA expression of α-FP was significantly reduced and the activities of ALP were remarkably declined(P0.05). Conclusions PA may inhibit proliferation and induce differentiation of human hepatocarcinoma SMMC-7221 cells.
