人外周血树突状细胞递呈可溶性肿瘤抗原触发抗瘤免疫的体外研究

摘　　要：考察人外周血树突状细胞（ＤＣ）递呈可溶性肿瘤抗原激发抗瘤免疫作用。应用改良Ｓｔｅｉｎｍ ａｎ 法分离纯化人外周血ＤＣ，同时将ＭＤＡ、３ＡＯ及Ｗｉｓｈ 三种瘤细胞制成可溶性肿瘤抗原，将二者于体外共育，分析ＤＣ表面标记的改变及某些细胞因子产生的水平，并检测其对肿瘤细胞杀伤作用的影响。结果表明，经可溶性肿瘤抗原刺激的ＤＣ分泌ＩＬ－１２及ＴＮＦα水平增加，同时高水平表达ＣＤ４０ 、ＣＤ８０ 等表面分子；与对照组相比，显示不同程度的促杀伤作用并有效地促进特异性淋巴细胞增殖。ＧＭ－ＣＳＦ上调ＤＣ的上述作用。Increasing evidence suggests that cell mediated immunity,especially T cell mediated immunity,is important for the control of tumor cells.Dendritic cells are potent professional antigen presenting cells and appear to process antigens for both MHC class Ⅰ and class Ⅱ pathways.In this study,we separated human dendritic cells from peripheral blood based on Steinman method with a slight modification and stimulated them with soluble tumor antigen(STA).The ability of dendritic cells in inducing tumor cell specific lysis was investigated.In addition,the surface marker of STA pulsed dendritic cells and their capability to secrete some cytokines were analysed.The results show that the IL 12 and TNFα secreted by STA stimulated denlrdtic cells were increased after culturing 48h in vitro and CD40,CD80 and MHC Ⅰ/Ⅱ membrane molecules were expressed in high level.It is necessary for dendritic cells to induce tumor cell specific lysis.GM CSF enhanced the effects of dendritic cells.These results suggest that using STA pulsed dendritic cells to stimulate anti tumor immunity may be a useful way of clinical tumor treatment.
