摘  要:目的:探讨双氢青蒿素对肺癌SPC-A-1细胞周期的阻滞作用及介导其周期阻滞的机制。方法:双氢青蒿素处理SPC-A-1细胞,PCR及免疫印迹检测生存素表达情况,基因转染建立生存素高表达SPC-A-1细胞,采用流式细胞术检测细胞周期变化。结果:双氢青蒿素可以诱导SPC-A-1细胞G2/M期周期阻滞,生存素表达下调,生存素高表达拮抗双氢青蒿素诱导的SPC-A-1细胞G2/M期周期阻滞。结论:生存素参与介导双氢青蒿素诱导的SPC-A-1细胞G2/M周期阻滞。Objective To explore the role of dihydroartemisinin (DHA) in cell cycle arrest in SPC-A-1 cells and the molecules associated with the cycle arrest. Methods SPC-A-1 (:ells were treated with DHA and the mRNA and protein expression levels of survivin were detected by PCR and Western blot. The cells with high level of survivin expression were established by genetic transfeetion. The changes in the cell cycle were measured by flow cytometry. Results DHA induced cycle arrest in G2/M in SPC-A-I cells, downregulating the expression of survivin. The cells with higher expression level of survivin resisted the inductive effect of DHA. Conclusions Survivin is involved in the process of G2/M arrest in SPC-A-1 cells induced by DHA.
