摘  要:为了探讨缺氧对体外培养在三维结构上的雪旺细胞的影响和EGb761 对缺氧雪旺细胞的保护作用,将在聚酯纤维上迁移的SD 大鼠的雪旺细胞进行缺氧培养,抗缺氧组培液含EGb761。正常组常规培养。结果显示,缺氧组:缺氧3 d,细胞停止迁移,细胞体由梭形变成半球形或球形;缺氧5 d,细胞开始死亡,从聚酯纤维上脱落。抗缺氧组:缺氧3 d 和正常组比较,细胞迁移和形态无明显改变;缺氧5 d,少数远离植块的细胞呈半球形和球形;缺氧7 d,少量死亡细胞脱离聚酯纤维。结果表明:缺氧将引起雪旺细胞迁移能力和方式及形态的改变,最后导致细胞死亡;EGb761 对缺氧的雪旺细胞有明显的保护作用。To observe the morphology and migration behavior of Schwann Cells(SCs) cultured on the three diamentional framework in hypoxia and the protection effects to the SCs against hypoxia by the Extract Gingkgo biloba 761(EGb761). The SCs obtained from sciatic nerve segments of newborn Sprague Dawley rats were cultured on the polyester fiber(PF). When a great number of SCs migrated along the PF, these cells were divided into three groups: Hypoxic group: cells cultured with the routine media without oxygen; Anti hypoxic group: cells cultured with the media contained EGb761 but without oxygen; Control group: cells didn't moved and the shape of cell changed in 3 div, many cells became swollen and curled up into a ball, collapsed, died and left away from the PF, Floated on the media nearby the PF, a little cells were seen on the PF while in 5 div; Anti hypoxic group: cells kept their primary speed and fashion of migration in 3 div, however, some cells changed its shape in 5 div, a few cells floted in the media, but a great number of cells migrated continuously as the normal group even in 7 div. Our experiments showed that the obviously different between these two kinds of groups and the EGb761 seemed effective in protecting the cultured SCs against hypoxia condition.
