摘 要:目的探讨大鼠脑缺血后不同时间轴突生长抑制因子Nogo-A在远隔部位小脑皮质的动态表达。方法 60只大鼠随机分为假手术组(n=30只)和大脑中动脉闭塞组(n=30只),每组分为1、3、5、7和10 d五个不同的亚组;采用改良的ZeaLonga法制备大鼠大脑中动脉闭塞模型;HE染色观察小脑皮质组织形态学改变;免疫组化染色检测小脑皮质Nogo-A蛋白的表达。结果 Nogo-A蛋白免疫反应性细胞广泛分布于小脑皮层少突胶质细胞(OLG)构成的髓鞘区域,主要位于OLG(胞体小、胞浆少和突起少)的细胞浆,并于脑缺血后第5 d、7 d和10 d较假手术组明显增高(P<0.05),第1 d和3 d无统计学差异(P>0.05)。结论大鼠大脑中动脉闭塞后远隔部位小脑皮层Nogo-A主要由OLG表达且呈动态变化。This study is aimed to explore the dynamic expression of Nogo-A protein in the remote area of cerebellum cortex after ischemia in rats at different times.We divided the rats into two groups randomly: sham operated group(n = 30) and middle cerebral artery occlusion(MCAO) model group(n = 30).The model of MCAO was constructed by adopting the upgrade of Zea Longa method.We employed HE and immunohistochemical techniques to detect the expression of Nogo-A protein.The result demonstrated that the immunopositive cells of Nogo-A protein main existed in the cytoplasm of cerebellar cortex's oligodendrocytes(small soma,few cytoplasm or protuberance).Compared to sum operated group,Nogo-A expression of MCAO was markedly increased at day 5,7 and 10 post-ischemia,(P〈0.05),but shown no statistical difference at day 1 and 3(P〉0.05).All the results implied that Nogo-A protein is main expressed by oligodendrocyte and demonstrates a dynamic change in the remote area of the opposite side cerebellum cortex of rats after MCAO.
