富血小板血浆在修复牙周组织缺损中的应用研究

摘　　要：目的:建立富血小板血浆(PRP)提取方法,以探讨作为自身复合生长因子来源的可行性,并研究其在修复牙周组织缺损中的作用。方法:利用二次离心法(1000 rpm×15 min;3000 rpm×8 min)分离全血,获得PRP。将PRP设3个浓度组(5%,10%,30%),比较各浓度PRP+植骨术与单纯植骨术治疗牙周骨内袋缺损的效果,以评价PRP在牙周组织再生治疗中的作用。结果:所获得的血小板浓度均超过全血的4倍,实验组各浓度的PRP+植骨术均可促进牙周组织的再生,实验组与对照组相比差异具有显著性(P<0.01)。当PRP浓度在5%～30%时促再生作用呈剂量依赖性。结论:二次离心法是一种简单易行的提取PRP的方法。PRP可促进牙周组织的再生。Objective：To establish a new method of isolating platelet-rich plasma（PRP） and the observation of its effects on the application in periodontal regeneration.Method：Platelet-rich plasma was isolated by two-step centrifugation（1000 rpm×15 min;3000 rpm×8 min） from 10 adults.Then the medium was dicarded and cells were exposed to 5 %,10 %,30 % PRP in 100 μL Alpha-MEM medium.To evaluate the effection of 5 %,10 %,30 % PRP and bone graft and bone graft only in periodontal regeneration.Result：This two-step centrifugation method sequestered and concentrated platelets to a level over four times baseline whole blood.PRP and bone graft can promoted periodontal regeneration.Conclusion：A convenient two-step centrifugation method of concentrating platelets was established.PRP produced by this technique and bone graft can promoted periodontal regeneration.At the dose of 5 %,10 %,30 % PRP promoted periodontal regeneration in a dose-dependant manner.
