脑胶质瘤中Jab1和p27的表达与意义

摘　　要：目的探讨Jab1和p27在脑胶质瘤中的表达、意义以及相互关系。方法运用免疫组化方法检测70例胶质瘤和8例对照脑组织标本中Jab1、p27蛋白的表达。Western blot检测6例新鲜胶质瘤标本中相应分子的表达。结果 Jab1在对照脑组织中表达不明显,在Ⅱ、Ⅲ、Ⅳ级胶质瘤中阳性率分别为21.2%、65.4%、91.7%,三者之间的表达水平差异有统计学意义(P<0.001)。p27在对照脑组织中表达明显,在Ⅱ、Ⅲ、Ⅳ级胶质瘤中阳性率分别为93.8%、53.8%、25.0%,三者之间的表达差异有统计学意义(P<0.001)。Western blot结果显示Jab1与p27在胶质瘤中的表达水平与肿瘤细胞的恶性程度相关。相关分析显示:胶质瘤中Jab1蛋白表达与增殖指标Ki-67表达呈正相关,与p27蛋白表达呈负相关;p27蛋白表达与Ki-67表达呈负相关(P<0.001)。结论胶质瘤中Jab1的异常高表达可能是抑制p27表达的重要因素。Jab1与p27的异常表达可能在胶质瘤的发生发展过程中起促进作用。Purpose To investigate the significance and the expression of Jab1 and p27 in the brain gliomas and their relationships.Methods The expression of Jab1 and p27 in 70 cases of human gliomas and 8 specimens of the normal brain tissue was detected by immunohistochemical technique.Western blotting was used to detect the expression of Jab1 and p27 in the fresh tumor tissues just removed from 6 gliomas.Results The expression of Jab1 protein was not obvious in all case of normal brain tissues.While in 70 cases of human gliomas,the positive rates of Jab1 expression in grades Ⅱ,Ⅲ,and Ⅳ were 21.2%,65.4%,and 91.7%,respectively.As for p27,the positive rates were 93.8%,53.8% and 25.0%,respectively.There were significant differences in the expression levels of both proteins among normal brain tissues,low and high grade gliomas（P0.001）.There was a positive correlation between Jab1 and Ki-67.Inverse correlation was found between protein of Jab1 and p27,also between p27 and Ki-67（P0.001）.Conclusion The over-expression of Jab1 in the gliomas may be an important factor for the inhibition of p27 expression.Abnormal expression of Jab1 and p27 may have promotive effects on the patholgenesis and development of the gliomas.
