摘  要:目的:观察降钙素基因相关肽(calcitonin gene-related peptide,CGRP)在坐骨神经分支选择性损伤(spared nerve injury,SNI)模型大鼠中央杏仁核中的表达变化。方法:健康SD大鼠分为假手术对照组(n=60)和坐骨神经分支选择性损伤组(n=60),实验组损伤后饲养3 d,7 d,14 d,免疫荧光染色结合图像分析技术观察CGRP在中央杏仁核中的表达变化。结果:CGRP分布于正常SD大鼠中央杏仁核,SNI后3 d,损伤组大鼠中央杏仁核中的CGRP表达开始升高,图像分析表明CGRP的光密度与对照组比较,差异有统计学意义(P<0.01);SNI后14 d CGRP表达达最高峰(P<0.01)。结论:SNI后CGRP可能通过杏仁核参与神经病理性疼痛的调控。Objective:To study the changes of calcitonin gene-related peptide(CGRP) expression in central nucleus amygdale in rat model of spared nerve injury.Methods:Healthy Sprague Dawley rats were randomly divided into sham operation group(n=60) and experimental group(n=60) which were killed at day 3,7 and 14 after spared nerve injury.The distribution and quantity of CGRP in central nucleus of amygdala was analyzed by immunofluorescence and image analysis methods.Results:CGRP-immunoreactivity(IR) existed in the central nucleuses of amygdale in normal Sprague Dawley rats.An initial increase of CGRP-IR was noted and the content of CGRP under image analysis was significantly higher compared with that of control group(P0.01) at day 3 after undergoing spared nerve injury and rise to a maximum level at day 14(P0.01).Conclusion:It was indicated that CGRP might play a role in the regulation of neuropathic pain after spared nerve injury.
