黄芪注射液对糖尿病大鼠肾脏BMP-7和CTGF表达的影响

摘　　要：目的观察黄芪注射液对糖尿病大鼠肾脏骨形成蛋白7(BMP-7)和结缔组织生长因子(CTGF)表达的影响。方法高脂高糖饲料喂养6周并注射STZ建立大鼠糖尿病模型,将成模的大鼠随机分为模型组和黄芪治疗组,并设正常对照组。各组大鼠干预8周。检测血糖(BG),计算肾质量/体质量(KI),放射免疫法检测24 h尿蛋白排泄率(UAER),全自动生化分析仪检测血总胆固醇(TC)、三酰甘油(TG),免疫组化法测定大鼠肾脏BMP-7和CT-GF蛋白表达情况,实时荧光定量PCR法检测大鼠肾脏BMP-7和CTGF mRNA表达情况。结果与正常对照组相比,模型组KI、BG、TC、TG、UAER显著增高,肾组织BMP-7蛋白和mRNA表达均明显下降,CTGF蛋白和mRNA表达均明显升高;与模型组比较,黄芪治疗组TC、TG、KI及24 h UAER显著降低,肾组织BMP-7蛋白及mRNA表达升高,而CTGF蛋白及mRNA表达明显下调。结论黄芪注射液对糖尿病大鼠肾脏有保护作用,其机制可能与恢复肾组织BMP-7表达,降低肾组织CTGF过度表达有关。Objective It is to observe the effect of astragalus injection on the expression of bone morphogenetic protein-7（BMP-7） and connective tissue growth factor（CTGF） in kidney tissues of rats with diabetes mellitus.Methods Rats model with diabetes mellitus were established with high carbonhydrate and high fat diet and intraperitoneal injection of streptozotocin（STZ）.The rats were randomly divided into model group and astragalus injection treated group.Meanwhile,a normal control group was set up.The rats in each group received corresponding treatment for 8 weeks.The urinary albumin excretion rate（UAER） was detected by radio-immunity method.The change of blood glucose were detected during the treatment period.TG and TC in blood were measured by automatic biochemistry analyzer.The expression of BMP-7 and CTGF mRNA in renal tissues were detected by RT-PCR.The expression of BMP-7 and CTGF protein in renal tissues were detected by immunohistochemistry assay.Results Compared with normal control group,blood glucose,TG,TC,kidney weight/body weight and UAER were increased significantly,the expression of BMP-7 protein and BMP-7 mRNA were decreased,the expression of CTGF protein and CTGF mRNA were increased significantly in model group compared with normal control group（P〈0.01）.However,in comparison with DM model group,the expression of BMP-7 and BMP-7 mRNA were increased and the expression of CTGF and CTGF mRNA were significantly decreased（P〈0.01 or P〈0.05） in astragalus injection treated group.Conclusion Astragalus injection has protecting function on kidney of rats with diabetic nephropathy,the mechanism of which may be related to up-regulating the expression of BMP-7 and down-regulating the expression of CTGF in renal tissues.
