胰岛素抵抗协同性激素刺激大鼠子宫平滑肌生长研究

摘　　要：目的:探讨胰岛素抵抗协同性激素刺激大鼠子宫平滑肌生长的作用,揭示胰岛素抵抗与子宫肌瘤形成间可能存在的关系。方法:将雌性、未孕、体重相似的Waster大鼠45只,随机分为3组:A组为对照组,B组为性激素刺激组,C组为性激素刺激+高糖高脂饲料饲养组,3组均饲养8周后测量体重、子宫重量,计算子宫脏器系数;测定空腹血糖、空腹血清胰岛素、血清雌二醇和孕酮水平,计算稳态模型胰岛素抵抗指数。子宫标本测量肌壁厚度,检测子宫平滑肌雌激素受体、孕激素受体和增殖细胞核抗原的表达。结果:B组HOMA-IR小于C组(P<0.05);B组E2小于C组(P<0.05);B组子宫脏器系数和子宫肌层厚度均小于C组(P<0.05),C组的ER、PR、PCNA表达均高于B组(P<0.05)。结论:胰岛素抵抗对性激素刺激大鼠子宫平滑肌生长具有促进作用,提示胰岛素抵抗有可能在子宫肌瘤的发生中具有作用。Objective： To observe the influence of insulin resistant to the growth of uterine smooth muscle by administrating female rats exogenous estrogen and progestogen,to reveal of the relation between insulin resistant and the formation of uterine leiomyomas.Methods： 45 female、non-pregnant、similar body weight Waster＇s rats were modeled,all rats were randomly divided into three groups：group A as normal control group,group B as animal model by exogenous sex hormone,group C as animal model by exogenous sex hormone plus insulin resistant group.All the animals were raised for 8 weeks,measurement of body weight and uterine weight,calculation the uterine coefficient;estimate fasting plasma glucose（FPG）、fasting serum insulin（FINS）、serum estrogen（E2） and serum progesterone（P）,calculation the HOMA-IR.Measurement the depth of myometrium,detected the expression of estrogen receptor（ER）,progesterone receptor（PR） and proliferating cell nuclear antigen（PCNA）.Results： HOMA-IR of group B were lower than that in group C（P〈0.05）;E2 of group B was lower that in group C（P〈0.05）;uterine coefficient and depth of myometrium of group B were lower than those in group C（P〈0.05）;the expression of ER、PR and PCNA of group C were higher than those in group B（P〈0.05）.Conclusion： There is a auxo-action of insulin resistant to the growth of uterine smooth muscle by administrating female rats exogenous estrogen and progestogen in this study,suspect that insulin resistant maybe has an influence to the formation of uterine leiomyomas.
