摘 要:目的以丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)为肝功能观察指标,探讨非酒精性脂肪性肝病(NAFLD)兔肝功能影响因素。方法 40只日本大耳白兔随机分为重度NAFLD模型组(重度组)、轻度NAFLD模型组(轻度组)、空白对照组(对照组)。重度组每只兔给予高脂饲料160 g/d,轻度组给予高脂饲料80 g/d+普通饲料80 g/d,对照组给予普通饲料160 g/d。饲养13周。饲养前耳中动脉采血5 mL于肝素管,离心、血浆保存于-20℃冰柜中。实验结束10%水合氯醛耳缘静脉麻醉,腹主动脉抽血5 mL于肝素管,离心、血浆保存于-20℃冰柜中。全部血浆按照设备、试剂操作常规检测甘油三酯、胆固醇、血糖、ALT、AST。在肝脏同一部位取1.5 cm×1.5 cm×1.5 cm组织1块,浸泡于10%甲醛固定液中固定,脱水石蜡包埋,切片,苏木素-伊红(HE)染色,光镜下观察肝脏组织形态学改变。结果①甘油三酯、胆固醇:饲养前重度组、轻度组、对照组甘油三酯、胆固醇比较差异无统计学意义(P>0.05)。饲养后甘油三酯、胆固醇重度组分别为(32.1±1.2)、(6.0±2.1)mmol/L,轻度组分别为(18.3±2.1)、(4.4±1.9)mmol/L,与饲养前比较差异有统计学意义(P<0.01);重度组高于轻度组,差异有统计学意义(P<0.01)。②血糖:饲养前重度组、轻度组、对照组血糖比较差异无统计学意义;饲养后重度组(11.0±1.2)mmol/L、轻度组(9.2±2.0)mmol/L与饲养前比较差异有统计学意义(P<0.01和<0.05);重度组高于轻度组,差异有统计学意义(P<0.05)。③ALT、AST:饲养前重度组、轻度组、对照组ALT、AST差异无统计学意义(P>0.05),饲养后重度组ALT(122±9)U/L、AST(126±9)U/L,轻度组ALT(83±7)U/L、AST(85±9)U/L,与对照组ALT(71±7)U/L、AST(72±4)U/L比较差异有统计学意义(P<0.01),重度组与轻度组比较差异有统计学意义(P<0.01)。④肝脏病理学:重度组重度NAFLD,轻度组轻度至中度NAFLD,对照组正常肝脏。结论 NAFLD兔�Objective To take the alanine amino transferase(ALT),the aspartic acid amino transferase(AST) as the liver function observation target and to investigate influencing factor of the liver function in the rabbit with non-alcoholic fatty liver disease.Methods Forty Japanese white rabbits were randomly divided into severe group with NAFLD(SG),mild group with NAFLD(MG),blank control group(BG).SG was given high fat diet 160 g/d,MG were given 80 g of high fat diet/day and normal diet 80 g/d,BG was given normal feed 160 g/d.The raising cycle is 13 weeks.5ml blood was drawn into heparin tube from ear middle artery before feeding,centrifuged and stored at-20 ℃to be tested.In the end of the experiment,10% chloral hydrate anesthesia was performed through the ear vein,5 mL blood was drawn into heparin tube from the abdominal aorta,centrifuged and stored at-20 ℃to be tested.According to the operation of routine testing,all samples were tested triglyceride(TG),cholesterol(TC),blood sugar(BS),ALT and AST.1.5 cm×1.5 cm×1.5 cm size of the liver tissue was sampled from the same position in the liver,stored in 10% formal dehyde fixation,dehydrated and embedded in paraffin,then sliced,HE stained and morphological changes of liver tissue were observed.Results ①TC,TG:For TC,TG of SG,MG and BG,there was no significant difference.After feeding,The TC,TG of SG is(32.1±1.2) and(6.0±2.1)mmol/L,MG is(18.3±2.1) and(4.4±1.9)mmol/L,respectively.After keeping SG,MG compared with the feeding before,there was a significant difference(P〈0.01).TG,TC of SG after feeding were higher than the MG,there was a significant difference(P〈0.01).②BS: SG of prefeeding,MG and BG have no significant difference in BS.After keeping SG(11.0±1.2)mmol/L and MG(9.2±2.0)mmol/L compared with the feeding before there was a significant difference(P〈0.01 and P〈0.05).BS of SG after feeding was higher than MG,there was a difference(P〈0.05).③ALT,AST:SG of prefeeding,MG and
