摘  要:目的和方法:实验观察亚硒酸钠对人急性髓性白血病细胞系HL- 60 细胞生长增殖、诱导分化、细胞内自由基产生和清除系统以及胞内环核苷酸累积的影响,探索亚硒酸钠对人白血病HL- 60 细胞作用机制。结果:8 ×106molL 或更高浓度的亚硒酸钠均可显著抑制HL- 60 细胞的生长增殖,作用5 d 可在一定程度上诱导HL- 60 细胞向成熟粒细胞方向分化。8 ×106 molL 亚硒酸钠作用一段时间,可增强自由基清除系统酶活性,减低胞内自由基水平,降低细胞内cGMP 累积而对cAMP 累积无明显影响。AIM and METHODS:To study the mechanism of antileukemic effect of sodium selenite, the effect of sodium selenite on the growth of HL-60 cells, its differentiation to mature granulocytic cells, activity of enzyme for free radical production and for their clearance, and accumulation of cyclic nucleotide were examined.RESULTS:8×10 6 mol/L-1.6×10 5 mol/L sodium selenite inhibite significantly the growth of HL-60 cells, being dose dependent. Exposure to 8×10 6 mol/L sodium selenite for 5 d induced differentiation of HL-60 cells in the direction of mature granulocytic cells to some extent. Exposure to 8×10 6 mol/L sodium selenite for 1~5 d increase activity of enzymes for clearing of free radical( indicated by SOD), decrease the content of free radical in cells (indicated by MDA) as time passes on. Exposure to 8×10 6 mol/L sodium selenite for 30 min has no effect on cAMP accumulation in cells either at rest or stimulated by adrenalin, but decreases cGMP accumulation and increases cAMP/cGMP ratio in the same time.CONCLUSION:It suggested that beside owing to a possible potential differentiation induction under certain circumstances, antileukemic effect of sodium selenite, may partially involve the increase of activity of enzymes for clearing free radical that cause decrease of free radical in cells.
