摘 要:目的研究乳腺癌中TSG101与MAPK/ERK信号通路的关系。方法应用脂质体法将靶向TSG101的siRNA转染入乳腺癌细胞株MCF-7中。分别使用免疫荧光和Western blot方法检测siRNA干扰后MCF-7中p-ERK蛋白表达的改变。使用免疫组织化学方法检测TSG101、p-ERK蛋白在54例人乳腺癌组织中的表达。结果 TSG101 siRNA转染后的MCF-7中p-ERK蛋白表达明显减少。TSG101和p-ERK蛋白在乳腺癌组织中的阳性率分别为74.07%、81.48%。TSG101和p-ERK在乳腺癌组织中的蛋白表达存在正相关(r=0.371,P<0.05)。结论 TSG101在乳腺癌中可能通过MAPK/ERK信号通路发挥生物学作用。Objective To investigate the relation between TSG101 and MAPK/ERK signal pathway in breast cancer. Methods TSG101 siRNA were transfected into breast cancer MCF-7 cell line via lipofection. The protein expressions of p- ERK in MCF-7 cells before and after transfection were assessed by and Western blot. Immtmohistochemical method was adopted to detect the expressions of TSG101 and p-ERK proteins in 54 cases of human breast cancer tissues. Results The expression of p-ERK protein was reduced significantly after TSG101 siRNA transfection than before. The positive expression rate of TSG101 and p-ERK was 74.07% and 81.48% separately in invasive duetal carcinoma of breast. The correlation between the expressions of TSG101 and p-ERK in breast cancer were significant. Conclusions The correlation between the expressions of TSG101 and p-ERK in breast cancer cells and tissues were significant. TSG101 may play biology effect through MAPK/ERK signal pathway in breast cancer.
