摘  要:目的:建立和评价肠易激综合征(IBS)大鼠模型,观察结肠肌间神经丛内神经元数目的变化,探讨其对肠易激综合征发病的意义。方法:采用乙酸灌肠和慢性激怒分别制造肠易激综合征大鼠模型,分别用腹壁肌电活动评估肠道敏感性及用结肠排便时间、排便颗粒评价肠道运动功能,免疫组化检测结肠肌间神经丛肠神经元标记物组织蛋白酶-D,计数各组肠神经元个数。结果:乙酸灌肠制造的IBS大鼠模型存在内脏感觉过敏,慢性激怒造成的IBS大鼠模型出现结肠运动功能紊乱。乙酸灌肠组肠神经元数目低于慢性激怒组和正常对照组(P<0.05),慢性激怒组与正常对照组肠神经元数目无显著差异(P>0.05)。结论:乙酸灌肠和慢性激怒方法制成的IBS动物模型可以用于IBS的实验研究。炎症因素致IBS大鼠模型结肠肌间神经丛存在神经元可塑性改变,推测这种改变可能是肠易激综合征发病的原因之一。Objective:To establish and evaluate rat model of irritable bowel syndrome,observe the changes of colonic myenteric and the number of neurons,and to explore the effect of changes in neuronal plasticity on irritable bowel syndrome.Methods:Rat model of irritable bowel syndrome was established by acetic acid enema and chronic irritation.The sensitivity of bowel was analyzed by testing abdominal electrical activity,and bowel motility was evaluated by defecation time and defecation particles.cathepsin-D was used to mark the neurons in the colonic myenteric plexus.Results:Models of irritable bowel syndrome by acetic acid enema showed visceral hypersensitivity,while models by chronic irritation had ccolonic motility disorder.The number of neurons in acetic acid enema group was significantly less than that of the chronic anger group(P〈0.05),while there was no significant difference between chronic anger group and control group(P〉0.05).Conclusions:Animal model of irritable bowel syndrome by acetic acid enema and chronic irritation both can be used in the experiment of irritable bowel syndrome.The missing of the neurons in the colonic myenteric plexus of irritable bowel syndrome induced by inflammatory factors may be one of the reasons for the pathogenesis of the irritable bowel syndrome
