摘 要:目的探讨银杏黄酮对颈淋巴阻断(CLB)后蛛网膜下腔出血(SAH)继发性脑损伤的保护作用。方法将Wistar大鼠随机分入对照组、SAH组、SAH+CLB组、SAH+CLB+生理盐水组、SAH+CLB+银杏黄酮50 mg组、SAH+CLB+银杏黄酮200 mg组,银杏黄酮经腹腔注射给予。制作CLB和SAH模型。于诱导SAH后3 d,用激光多普勒血流计测定局部脑血流量(rCBF),干湿重比较法测定脑组织含水率,免疫荧光法检测血红素氧合酶-1(HO-1)表达。结果 SAH后3 d rCBF显著降低,CLB可导致rCBF进一步下降,银杏黄酮可减轻rCBF下降的程度;SAH导致脑组织含水率升高,并在CLB时进一步增加,银杏黄酮对之具有拮抗作用;SAH促进脑组织HO-1表达,CLB后HO-1表达进一步增加,银杏黄酮促进CLB后SAH大鼠脑组织HO-1表达。结论 CLB可加重SAH后脑损伤,银杏黄酮对之具有一定缓解作用。OBJECTIVE To investigate the protective effect of ginkgetin, a component of the extract of Ginkgo biloba, on cerebral injury following subarachnoid hemorrhage(SAH) after cervical lymphatic blockage (CLB). METHODS Wistar rats were divided into normal control, SAH, SAH + CLB, SAH + CLB + (Normal saline, NS) , SAH + CLB + Ginkgetin 50 rag, and SAH + CLB + Ginkgetin 200 mg groups. Ginkgetin was given by intraperitoneal injection. CLB models were developed by .removal of cervical lymphatic nodes 'after obstruction of the tubes, and SAH models were established by double cisternal injections of autologus arterial blaod, On the third day after the second cisternal injection, regional cerebral blood flow (rCBF) was detected using a laser Doppler flowmeter, A dry and wet weight comparison method was used to calculate brain water content. The expression of heine oxygenase-1 in brain section was determined by an immunofluorescence assay. RESULTS SAH led to decreased rCBF which was deteriorated by CLB, An increase in brain water content was observed after SAH and it was aggravated by CLB. The changes were antagonized by the use of Ginkgetin. An increased expression of HO-1 in brain sections was noted after SAH. CLB led to further increase of HO-1 expression, The application of Ginkgetin augmented the HO-1 expression. CONCLUSION CLB deteriorates cerebral injury after SAII which may be partly reversed by the use of Ginkgetin.
