摘 要:比较基因组杂交是近年来在荧光原位杂交基础上发展起来的一种新的分子遗传学方法,具有一次实验可在全部染色体或染色体亚区水平上对整个基因组内 D N A 物质的增加或减少进行分析,且标本来源不受限制、不需进行肿瘤细胞培养等优点,已广泛用于各种肿瘤和遗传性疾病的检测。本文较详细地介绍了该项技术的基本原理和具体操作方法以及在肿瘤临床中的应用,并对其优缺点及应用前景进行了评价。Comparative genomic hybridization (CGH) is a new molecular cytogenetic method developed recently on the basis of fluorescence in situ hybridization(FISH). It has such many advantages as the gain or loss of whole genomic DNA at the levels of total chromosomes and sub chromosomes can be analyzed in an experiment, with no restrictions on source of specimens and no need for culture cancer cells and so on. It has been widely used in the detection of various types of tumors and hereditary diseases. The basic principles and specific procedures of CGH as well as applications to clinical tumors were introduced,and both advantages and shortages were assessed.
