摘 要:目的检测纺锤体检测点蛋白Bub1和Mad2在煤焦沥青致人支气管上皮细胞系(BEAS-2B)癌变细胞中的变化,探讨Bub1和Mad2在煤焦沥青接触者肺癌发生中的作用。方法利用中温煤焦沥青烟提取物作为诱导剂,建立永生化BEAS-2B的恶性转化模型,用实时荧光定量逆转录-聚合酶链反应(RT-PCR)检测Bub1和Mad2基因的mRNA表达水平;采用细胞爬片免疫组化半定量法检测蛋白表达改变。结果与正常对照组和二甲基亚砜(DMSO)组相比较,煤焦沥青烟提取物诱导转化细胞Bub1和Mad2基因mRNA和蛋白表达水平均减低,差异具有统计学意义(P<0.05)。正常对照组和DMSO组之间2种基因的mRNA和蛋白表达水平均没有明显改变,差异无统计学意义(P>0.05)。结论在煤焦沥青致支气管上皮细胞恶性转化后,纺锤体检测点蛋白Bub1和Mad2表达水平降低。Objective To detect the changes of spindle checkpoint protein Bub1 and Mad2 in malignant transformation of BEAS-2B cells induced by coal tar pitch smoke extracts,and to study the role of Bub1 and Mad2 in the nascency of lung cancer.Methods BEAS-2B cells were induced by coal tar pitch smoke extracts to form malignant transformation in vitro.The Bub1 and Mad2 expression levels of mRNA were assessed by real-time reverse transcription polymerase chain reaction(RT-PCR),the protein expression variation was determined by cell culture overslip of immunohistochemical methods.Results Compared to the negative control group and DMSO control group,mRNA of Bub1 and Mad2 and protein expression levels of the extract group was decreased(P〈0.05),while there was no significant difference between the control groups(P〉0.05).Conclusion The expression levels of Bub1 and Mad2 decreased after the malignant transformation in bronchial epithelial cells BEAS-2B induced by coal tar pitch smoke extracts.
