摘  要:目的:观察通阳药物葱白提取物对肝窦毛细血管化(HSC)模型大鼠肝组织基质金属蛋白酶及其抑制因子MMP-2、TIMP-2蛋白及mRNA表达的影响,并初步探讨其作用机理。方法:SD大鼠经酒脂混合物灌胃111天后,用40%CCl40.3 mL/100 g连续4周皮下注射造成HSC模型。皮下注射的同时予葱白提取物治疗19周。采用Western-Blotting方法和RT-PCR方法检测各组大鼠肝组织MMP-2、TIMP-2蛋白及mRNA的表达。结果:(1)葱白提取物组大鼠肝组织MMP-2、TIMP-2蛋白的表达量较模型组有均显著降低(P<0.01)。(2)葱白提取物组大鼠肝组织MMP-2mRNA、TIMP-2mRNA的表达量较模型组均有显著降低(P<0.01)。结论:通阳药物葱白提取物通过促进MMP-2/TIMP-2的表达平衡而起到促进阳气条达的作用。Objective:To study the effects and its mechanism of fistular onion stalk extraction on hepatic sinusoid capillarizatin(HSC)rats by observing the changes of Protein Mmp-2,Timp-2 and Mmp-2mRNA,Timp-2mRNA in livers of model rats.Methods SD male rats were fed the mixture of alcohol and fat for 111 days.And then were given a hypodermic injection of 40% CCL4 0.3 mL/100g in the subcutaneous tissue of legs for 4 weeks continually.Fed fistular onion stalk extraction at the same time of injection for 19 weeks.Western-blotting and RT-PCR was used to assay the levels of Protein Mmp-2、Timp-2 and Mmp-2mRNA、Timp-2mRNA in the livers of rats.Results:Results(1)The expression of MMP-2、TIMP-2 in Fistular Onion Stalk Extraction both expressed lower than which in model group(P0.01).(2)The expression of MMP-2mRNA、TIMP-2mRNA in Fistular Onion Stalk Extraction expressed lower than which in model group(P0.01).Conclusion:Fistular onion stalk extraction can make Yang Qi circulate fluently by encouraging to establish a new balance of MMP-2/TIMP-2 in the model rats of HSC.
