摘 要:目的建立苏氏接骨胶囊中当归和川芎的定性鉴别和总阿魏酸的定量检测方法。方法采用薄层色谱法对苏氏接骨胶囊中的当归和川芎进行定性鉴别,采用高效液相色谱法对阿魏酸进行定量测定。色谱柱为Thermo C18(250mm×4.6mm,5μm)柱,流动相为乙腈-1%醋酸溶液(20:80),体积流量1.0mL/min,柱温25℃,检测波长320nm。结果薄层色谱清晰,分离良好,阴性无干扰。阿魏酸在0.42~2.24μg线性关系良好(r=0.9999),平均回收率为99.26%,RSD为1.27%。结论所建立的方法准确、快速、简便、重现性好,可作为苏氏接骨胶囊的质量控制方法。Objective To establish methods for identification and quantification of Angelica sinensis and Ligusticum chuanxiong in Su’s Fracture Healing Capsule.Methods Su’s Fracture Healing Capsules were identified by TLC.A new HPLC method was developed to quantitatively analyze the total ferulic acid which was the sum of free ferulic acid and hydrolyzed components.A Thermo C18(250 mm×4.6 mm,5 μm)was used with acetonitrile-1% acetic acid(20:80)as the mobile phase.The flow rate was 1.0 mL/min and the column temperature was at 25 ℃.The detective wavelength was set at 320 nm.Results Spots obtained from the test solutions had the same color as Angelica sinensis and Ligusticum chuanxiong reference substances and the blank solution had no interference.The linearity range of ferulic acid was 0.42—2.24 μg.The average recovery of ferulic acid was 99.26% with the RSD of 1.27%.Conclusion The method of identification and quantification were accurate,simple and reproducible.It can be used for the quality control of Su’s Fracture Healing Capsule.
