抑癌基因RASSF1A与RASSF1B基因的关系

摘　　要：目的:研究RASSF1基因2个转录本基因在胃癌组织及相对应的癌旁组织中表达情况以及RASSF1 2个转录本基因与肿瘤发生的相关性。方法:设计特异性的引物,用逆转录-聚合酶链反应(RT-PCR)的方法检测胃癌及癌旁组织中RASSF1家族中2种不同转录本mRNA的表达情况。结果:40例胃癌组织中RASSF1A基因表达缺失率为60.2%(24/40),RASSF1B基因表达缺失率为37.5%(15/40)。结论:RASSF1A和RASSF1B2种转录本在胃癌组织中的表达下调,RASSF1A基因在胃癌的发展中起重要的抑制作用,RASSF1B的表达还可能与细胞的分化程度有关。Objective： To examine the expression of ras-association domain family 1（RASSF1） gene in gastric carcinomas tissues and analyze the relationship between transcripts of RASSF1 gene and carcinogenesis.Methods： The expression of RASSF1 gene in Gastric Carcinomas Tissues was examined by RT-PCR with anchored primers.Results： The rates of expression loss of transcript A and transcript B were 60.2%（24/40） and 37.5%（15/40）,respectively.Conclusions： The expression of RASSF1 gene isoforms is down-regulated in gastric cancer tissues,which is negative correlation to cancer,RASSF1A gene is important during the developmeng of the Gastric cancer.It is suggested that transcript B may be candidate genes for inhibiting cancers.Relations of them are not obviours.
