摘 要:目的:探讨龙芽楤木皂苷(As)对乙醇所致小鼠急性肝损伤的保护作用及其机制。方法:采用乙醇灌胃法制备小鼠急性乙醇性肝损伤模型。50只雄性昆明小鼠随机分为对照组、急性乙醇性肝损伤模型组、As低剂量干预组(30mg.kg-1)和As高剂量干预组(60mg.kg-1)。比较各组小鼠肝脏指数、血清丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)活性及肝组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、丙二醛(MDA)含量及肝组织病理形态变化。结果:10d时急性乙醇性肝损伤小鼠肝组织SOD、GSH-Px活性降低(P<0.01),血清ALT、AST活性、肝脏脂质及肝组织MDA含量增加(P<0.05),与对照组比较有显著差异。给予不同剂量As后,急性乙醇性肝损伤小鼠肝组织SOD、GSH-Px活性均升高(P<0.01),血清ALT、AST活性和肝组织MDA含量降低(P<0.05),肝脏组织病理损伤不同程度地改善。结论:As能有效地改善肝功能、减轻脂质过氧化反应,对乙醇所致大鼠急性肝损伤具有预防作用。Objective:To observe the prevention of Aralosides(As) originated from Aralia Elata Miq Seem on acute alcoholic liver injury in mice.Methods:A total of 50 male mice were randomly divided into 4 groups: control group(Group N),model group(Group M),As low dose treated group(Group L) and As high dose treated group(Group H).On day 10 all mice were sacrificed.Liver pathological changes were examined with the activities of serum alanine aminotranferease(ALT),aspartate aminotranferease(AST),the activities of liver superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) and the level of liver malondia-ldehyde(MDA).Results:At the end of 10 days,the activities of serum ALT,AST and the level of liver MDA were significantly increased in model group compared with those of control group(P0.05).The activities of SOD and GSH-Px were highly decreased in model group compared with those of control group(P0.01).After given different dose of As,the activities of serum ALT,AST and the level of liver MDA were significantly decreased compared with those of model group(P0.05).Meanwhile,the activities of SOD and GSH-Px were highly increased compared with those of model group(P0.01).There were typical liver pathological changes observed in the two As groups.Conclusions:As can effectively prevent acute alcoholic liver injury,which may be due to its effects in prevention of lipid peroxidation.
