摘  要:目的研究RhoC基因对胆管癌QBC939细胞株的增殖及cyclinD1 mRNA表达的影响。方法将正、反义RhoC cDNA真核表达载体,转染人胆管癌细胞QBC939,检测细胞生长曲线,RT-PCR检测cyclinD1在QBC939细胞、转染空载体的QBC939细胞(QBC939-V)、转染正义RhoC的QBC939细胞(QBC939-S)、转染反义RhoC的QBC939细胞(QBC939-AS)的mRNA相对表达量。结果与QBC939及QBC939-V对照细胞相比,QBC939-S体外生长较快,QBC939-AS体外生长较慢,cyclinD1表达在QBC939-AS与对照组减少(P<0.05),cyclinD1表达在QBC939-S与对照组增加(P<0.05)。结论 RhoC可能通过调节cyclinD1来调控QBC939的增殖能力。Objective To observe the effect of RhoC gene on QBC939 cells' proliferation and cyclinD1 gene expression. Methods Sense,Antisence eokaryotie expression plasmid of RhoC were transfected into QBC939 by Lipofectamine method. RT - PCR was used to examine the expression of Hygromyein B and cyclinD1, the changes of proliferation were checked by growth analysis in vitro. Results The difference of eyelinD1 expression in QBC939 - AS group, QBC939 - S group and in control group was statistically significant( P 〈0.05 ), Compared with the control QBC939 cells and QBC939 -V cells, QBC939 - AS grew more slowly, but QBC939 - S grew more quickly. Conclusion RhoC could control the proliferation of eholangioearcinoma cell lines QBC939 cells by control the expression of cyclinDl.
