海豹油对酒精性肝病预防作用的实验研究

摘　　要：目的:观察海豹油对酒精性肝病的预防作用。方法:采用酒精灌胃制备酒精性肝病大鼠模型。Wistar雄性大鼠60只,随机分为6组,分别为肝损伤模型对照组、空白对照组、阳性对照组及海豹油的低、中、高三个剂量组。各剂量组每天经口灌胃给予海豹油,空白对照组给予蒸馏水,模型对照组给予色拉油,阳性对照组给予海王牌金樽片。于实验的第30天各组大鼠一次性经口给予50%乙醇12 ml/kg(空白对照组动物给蒸馏水),禁食16 h处死动物,采血,剖腹取肝,制备肝匀浆。测定血清中的天冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(AIT)、甘油三脂(TG)、胆固醇(TC)和肝组织中的丙二醛(MDA)、谷光甘肽(GSH)、超氧化物歧化酶(SOD)和TG含量并进行病理组织学检查。结果:与模型对照组比较,海豹油3.0 ml/kg剂量组能明显降低血清ALT和肝组织MDA含量,海豹油3.0 ml/kg、6.0 ml/kg剂量组能明显降低酒精性肝病大鼠血清AST、TG、TC含量和肝组织中TG含量,增强SOD酶的活性,并能改善肝细胞脂肪变性。结论:海豹油对大鼠酒精性肝病有预防作用。Objective：To study the preventive effects of seal oil on alcohol liver disease of rats.Methods： Wistar male rats were given alcohol intragastrically to prepare rat model of alchol liver disease（ALD）.60 rats were randomly divided into 6 groups,respectively,the model group,blank control group,positive control group and the low,medium and high dose group of seal oil.Each dose group were orally administered daily for seal oil,blank control group and model control group was given distilled water and salad oil respectively,positive control group was given the Hai Wang Jin zun tablet.After 30 days of the experiment,rats were were orally administered 50% ethanol 12 ml/kg in model group,positive control group and seal oil group,rats were orally administered distilled water in blank control group.After 16h fasting,all rats were sacrificed,taken blood,,removed liver and preparation of liver homogenate.The effects of seal oil on alchol liver disease was assessed based on the evaluation of pathology,bio-chemical parameters of serum and liver homogenate.Results： Compared With the model control group,the 3.0 ml/kg seal oil group could decreased serum ALT and MDA concentration in liver of ALD rats,the 3.0 ml/kg～6.0 ml/kg seal oil group could reduced serum AST,TG,TC and TG content in liver tissuin,enhanced the activity of SOD significantly.Pathologic histomorphology investigation showed that seal oil markedly improved liver steatosis.Conclusion： Seal oil can protect liver cells from acutelv damaged by alchol and prevent ALD significantly.
