复方中药紫龙金抑制肾癌ketr-3细胞增殖的血清药理学研究

摘　　要：目的观察复方中药紫龙金含药血清对人肾癌ketr-3细胞生长增殖的抑制作用。方法运用血清药理学方法,选择不同浓度的含紫龙金大鼠血清与ketr-3细胞共同孵育,于不同时间用生物倒置显微镜观察细胞形态变化,W ST-8比色法检测紫龙金含药血清对人肾癌ketr-3细胞的增殖抑制率。结果紫龙金含药血清作用ketr-3细胞48 h后,细胞呈现典型的凋亡形态学改变,可见部分细胞变圆,体积变小,有凋亡小体产生;且抑制作用呈现时间和剂量依赖关系,与对照组比较,各浓度含药血清(5%、10%、20%)作用24 h后细胞增殖抑制率无明显差异(P>0.05),作用48 h后细胞增殖抑制率分别为4.9%(P>0.05)、22.6%(P<0.01)、28.7%(P<0.01)。结论紫龙金含药血清可有效抑制人肾癌ketr-3细胞体外增殖。Objective To investigate the inhibitory effects of serum with Zilongjin on human renal carcinoma cell line ketr-3.Methods By means of serum pharmacologic method,medicated serum samples containing Zilongjin of different concentrations and human renal carcinoma cell line ketr-3 were co-incubated to observe the morphological changes of the cells at different time points with biological inverted microscopy,and WST-8 assay was used to detect the inhibition of ketr-3 cell proliferation.Results 48 hours after treatment with different concentrations of medicated serum containting Zilongjin,the ketr-3 cells exhibited typical morphologic changes of apoptosis,including evident round shape in some cells,cell shrinkage and appearance of apoptotic bodies;Zilongjin serum could cause inhibition on the proliferation of ketr-3 cells in time-dependent and dose-dependent manners.Compared with the control group,there was no significant difference in the inhibition of cell proliferation for each concentration of medicated serum（5%,10% and 20%） at 24 h（P0.05）.At 48 h,the inhibition rate of cell proliferation was 4.9%（P0.05）,22.6%（P0.01） and 28.7%（P0.01）,respectively.Conclusion Zilongjin-medicated serum can effectively inhibit the proliferation of human renal carcinoma cell line ketr-3 in vitro.
