Tim-1及Tim-3mRNA在胃癌患者外周血单个核细胞中的表达及临床意义

摘　　要：目的研究T细胞免疫球蛋白黏蛋白1（Tim-1）及T细胞免疫球蛋白黏蛋白3（Tim-3）mRNA在胃癌患者体内的表达与其临床意义。方法采用RT—PCR法分别检测22例胃癌患者及8例正常人外周血单个核细胞中Tim-1及Tim-3mRNA的表达情况，并分析它们与临床各病理参数之间的关系。结果Tim-1mRNA在胃癌中的表达高于正常对照组[（11．38±1．85）比（9．64±1．53）]，差异有统计学意义（P〈0．05），且与肿瘤浸润深度、分化程度，淋巴结转移、临床分期有关（P〈0．05），Tim～3mRNA在胃癌中的表达高于正常对照组[（11．74±1．71）比（9．81±1．69）]，差异有统计学意义（P〈0．05），但Tim-3mRNA在胃癌中的表达与各病理参数无关（P〉0．05）。结论Tim-1及Tim-3mRNA在胃癌患者中的表达均高于正常对照组，Tim-1mRNA在胃癌患者中的表达还与肿瘤的侵袭转移密切相关，Tim-1及Tim-3有可能成为胃癌基因治疗的新靶点。Objective To explore the expression of Tim - 1 and Tim - 3 mRNA in PBMCs from gastric carcinoma patients and its clinical significance. Methods The expression of Tim - 1 and Tim -3 mRNA was detected by fluorescent quantitative real time RT - PCR in 22 gastric carcinoma patients and 8 normal subjects. The association of the expressions with clinical - pathological indicators was examined. Results The expression of Tim - 1 in the PBMCs from gastric carcinoma patients was higher than that from normal controls （ 11.38 ± 1.85 vs 9.64 ± 1.53） （ P 〈 0. 05 ）. The expression of Tim - 1 was closely related to the depth of invasion, tumor differentiation, lymph node metastasis and TNM stages （P 〈 0.05 ）. The expression of Tim - 3 in the PBMCs from gastric carcinoma patients was higher than that from normal controls （ 11.74 ± 1.71 vs 9.81 ± 1.69） （P 〈 0.05 ）, but the expression of Tim - 3 showed no close relationship with the clinical - pathological indicators. Conclusion Tim - 1 and Tim - 3 may be involved in the occurrence and development of gastric cancer and become new targets for gene therapy of gastric cancer.
