摘  要:目的:研究重组人钠/碘同向转运体基因(hNIS)腺病毒介导肺癌A549细胞的碘摄取、外流和杀伤情况,为放射性碘治疗肺癌提供理论和实验依据。方法:用重组hNIS腺病毒(AdNIS)感染肺癌A549细胞,检测感染细胞内hNIS蛋白的表达;在体外培养的条件下研究其125I的摄取、外流情况以及131I的杀伤情况。结果:hNIS蛋白在实验组肺癌A549细胞中表达阳性且主要分布于细胞膜上;实验组细胞摄碘能力较对照组高;细胞摄碘后碘外流过程十分迅速;实验组细胞可被131I有效杀伤。结论:应用腺病毒为载体,可以有效的将hNIS基因转染至体外培养的肺癌A549细胞中,并介导肺癌细胞对放射性碘的摄取及选择性杀伤作用。Objective:To explore the radiotherapy of lung cancer by using an adenoviral vector to deliver the hNIS gene into the A549 cells.Methods:By culturing the A549 cells in vitro and detecting the expression products of hNIS gene,the uptake and efflux of iodide was examined.In addition,the cells were exposed in 131I to assess the efficiency of this cancer gene therapy strategy.Results: Immunohistochemistry assay showed the expression of hNIS protein was positive in the experimental group and hNIS proteins were detected in the cell membranes.Infection of A549 cells with AdNIS resulted in sensitive 125I uptake by these cells to a level 10 times higher than that in the noninfected cells.The efflux of iodide was also rapid.AdNIS-infected tumor cells were selectively killed by exposure to 131I.Conclusion: The hNIS gene can be transfected to A549 cells mediated by adenoviral vector.The AdNIS leads to efficient iodide uptake in vivo but the efflux of iodide was also rapid.AdNIS-infected tumor cells were selectively killed by exposure to 131I.
