摘  要:目的探讨塞来昔布对LoVo移植瘤中环氧化酶-2(COX-2)、血管内皮生长因子(VEGF-C)的表达及对微血管生成的影响。方法移植LoVo的裸鼠分为三组,分别予安慰剂、塞来昔布40 mg/kg和80 mg/kg,第21天取瘤,测定抑瘤率和相对肿瘤增殖率,免疫组化法检测移植瘤中COX-2和VEGF-C的表达,并测定肿瘤微血管密度(MVD)。结果①VEGF-C的表达与COX-2的表达相关,三组中COX-2和VEGF-C的表达差异无统计学意义;②MVD与VEGF-C、COX-2表达相关,给药组MVD低于对照组。结论 COX-2和VEGF-C的表达与肿瘤血管生成有密切关系;塞来昔布可抑制肿瘤新生血管生成。Objective To investigate the effect of celebrex on the expression of COX-2 and VEGF-C in the LoVo xenograft,and to study the effect of celebrex on angiogenesis.Methods The transplantable human colon carcinoma tumor models in nude mice were established and were divided into 3 groups at random,and treated with placebo,celebrex 40 mg/kg,celebrex 80 mg/kg,respectively.Mice were sacrified at 21th day.Count the tumor inhibition rate and relative tumor proliferation rate.The expressions of COX-2,VEGF-C and the microvessel density(MVD),were detected by immunohistochemical staining.Results ①The expression of COX-2 and VEGF-C was correlated,but there was no difference among the three groups.②MVD was correlated with the expression of COX-2 and VEGF-C,the level of MVD in the groups with celebrex was lower than the controls.Conclusion The expression of COX-2 and VEGF-C are correlated with the angiogenesis of the transplant tumor.Celebrex can inhibit the angiogenesis of the transplant tumor.
