摘  要:目的探讨声动力化学疗法(SDT)对大鼠脑胶质瘤凋亡及相关基因表达的影响。方法以大鼠颅内C6胶质瘤为实验模型,观察SDT组、单纯照射组、血卟啉单甲醚(HMME)组、对照组脑胶质瘤组织学变化、原位凋亡检测、Cyt-C和Caspase-3蛋白表达水平。结果 SDT组随着时间的延长,肿瘤组织坏死逐渐增加,24h达到高峰;单纯照射组可见肿瘤组织坏死较SDT组明显少;对照组及HMME组标本各时间段胶质瘤细胞均未见坏死组织。原位凋亡检查见HMME组和对照组凋亡细胞极少,各时间点凋亡率无明显差别。处理后24h,单纯照射组凋亡率明显高于HMME组和对照组(P<0.01);处理后3、7d,凋亡率与HMME组和对照组无明显差别。SDT组处理后24h,凋亡率明显高于其他3组(P<0.01)。处理后3、7d,凋亡率与其他3组无明显差别。HMME组和对照组各时间点Cyt-C和Caspase-3蛋白表达率无明显差别。单纯照射组Cyt-C和Caspase-3蛋白表达率高于HMME组和对照组(P<0.01);处理3、7d后Cyt-C和Caspase-3蛋白表达率与HMME组和对照组无明显差别。SDT组处理后24h,Cyt-C和Caspase-3蛋白表达率明显高于其他3组(P<0.01)。处理后3、7d,Cyt-C和Caspase-3蛋白表达表达率与其他3组无明显差别。结论 SDT对大鼠脑胶质瘤有明显的抑制效应,早期可能通过损伤线粒体后,释放Cyto-C,激活Caspase-3而直接启动胶质瘤细胞本身凋亡程序。Objective To study the apoptosis and related gene expression of SDT on C6 glioma cells in vitro.Methods The rat brain C6 glioma was as experimental model to observe process changes of brain glioma in situ apoptosis detection,Cyt-C protein,Caspase-3 protein expression level of four(SDT,irradiation,HMME,control) groups.Results With time,SDT group had the increased tumor necrosis,24 h reached the peak;irradiation group had visible tumor necrosis,but significantly less than that of SDT group;control group and each time group had HMME glioma cells without necrotic tissue.Apoptosis in situ inspection showed control group and HMME group had few apoptotic cells at different time points without significant difference.Treated for 24 h,apoptotic rate of irradiation group was significantly higher than that of HMME and control groups(P0.01);treated for 3,7 d,had no significant difference.Treated for 24 h,apoptosis of SDT group was significantly higher than that of the other groups(P0.01);treated for 3,7 d,had no significant difference.Cyt-C and Caspase-3 protein expression results showed,HMME and control groups at different time points had no significant difference;Irradiation group treated 24 h,proteins expression were higher than those of HMME and control groups(P0.01);treated for 3,7 d,expression rates had no significant difference.SDT group treated for 24 h,proteins expression was significantly higher than that of the other groups(P0.01).Treated for 3,7 d,protein expression had no significant difference among the other three groups.Conclusions SDT has significant inhibitory effect on rat glioma,possibly through damaging mitochondria early after the release of Cyto-C,activation of Caspase-3,starting directly apoptosis process of glioma cells.
