摘  要:目的:探讨核转录因子(nuclear factor kappa B,NF-κB)抑制剂吡咯烷二硫代氨基甲酸盐(pyrrolidine dithioearba-mate,PDTC)对脂多糖(lipopolysaccharide,LPS)刺激的人牙周韧带细胞的干预作用。方法:采用组织块法分离培养人牙周韧带细胞(human periodontal ligament cells,HPDLCs)。实验分为对照组与实验组,对照组细胞以含10μg/ml LPS的培养基培养;实验组分别以PDTC10、100、1 000μmol/L预处理细胞30 min后,加入含相同浓度LPS的培养基培养。ELISA检测细胞上清液中炎性细胞因子IL-1β、IL-8分泌量的改变。免疫荧光检测干预前后细胞内NF-κB的核转移改变。结果:PDTC抑制HPDLCs增殖,10μmol/L PDTC预处理对HPDLCs分泌IL-1β和IL-8无影响,其余2种浓度均可抑制IL-1β和IL-8的分泌,1 000μmol/L PDTC的抑制作用更强,与其它各组相比差异具有显著性(P<0.05)。免疫荧光显示PDTC预处理后细胞核NF-κB的表达明显减弱或消失。结论:PDTC可以抑制LPS刺激的HPDLCs分泌IL-1β与IL-8和细胞内NF-κB的核转移,说明NF-κB信号通路在牙周病的发生过程中具有重要作用,阻断该通路可能成为牙周病药物治疗的一种新策略。Objective:To study the effects of pyrrolidine dithioearbamate(PDTC),a NF-κB inhibitor,on lipopolysaccharide(LPS)-stimulated human periodontal ligament cells(HPDLCs).Methods:HPDLCs were prepared by using explant culture technique.Cells were devided into two groups.Control group was treated with culture medium containing 10 μg/ml LPS.Experimental group was incubated with 10,100 and 1 000 μmol/L of PDTC for 30 min respectively before culture medium containing 10 μg/ml LPS was added.Then IL-1β and IL-8 secreted into the culture medium was determined by an enzyme-linked immunosorbent assay(ELISA).Nuclear translocation of NF-κB in the cells was observed by indirect immunofluorescence.Results:PDTC suppressed HPDLCs proliferation.10 μmol/ml PDTC didn' t inhibit the secretion of IL-1β and IL-8,but 100 μmol/L and 1 000 μmol/L PDTC did.And,more pronounced inhibition of 1 000 μmol/L PDTC showed that there was significant difference compared with the others(P0.05).The NF-κB antibody staining showed strong green fluorescence signals in the nuclei after LPS stimulated,but PDTC treatment strongly reduced the intensity of nuclear staining.Conclusion:IL-1β and IL-8 expression and nuclear translocation of NF-κB in LPS-stimulated HPDLCs can be inhibited by PDTC.NF-κB signaling pathway play a role in the development of periodontal disease.
