摘 要:目的利用人胃癌细胞SGC-7901建立裸鼠胃癌腹膜转移模型。通过基因芯片技术筛选出与胃癌腹膜转移相关的所有基因,探讨胃癌腹膜转移的机制。方法体外培养人胃癌细胞株SCG-7901细胞,并以此建立胃癌裸鼠原位移植瘤模型;收集原发灶和转移灶标本,提取RNA,制备探针,进行基因芯片杂交找出差异基因。结果共建立供实验所需的原位移植裸鼠模型15只。胃壁成瘤率为100%(15/15),发生腹膜转移率为40%(6/15)。通过芯片杂交数据分析结果,计算Ratio(Cy3/Cy5,即转移灶/原发灶值)。确定差异基因筛选标准为:Ratio≥2为上调基因,Ratio≤0.5为下调基因。实验发现192个上调基因和139个下调基因。结论利用基因芯片技术筛选出数百个胃癌腹膜转移的差异表达基因。上调基因在胃癌腹膜转移中起到了促进作用,而下调基因在胃癌腹膜转移中起到了抑制作用。实验结果为研究与胃癌腹膜转移的相关基因提供了依据与参考。Objective To develop the animal model with peritoneal metastasis of human gastric cancer by SGC-7901. To search differential expression gene in peritoneal metastasis of gastric cancer by gene chip and to study the mechanism of human gastric cancer metastasis. Methods SGC-7901 cells were cultured in RPMI- 1640 medium. To establish a metastasis model of human gastric cancer metastasize to intraperitoneal cavity by SGC-7901 cells. Extract the total RNA from the gastric cancer cell and gastric cancer metastasize cell, analysed the differentially expressed genes which were recoursed in the gene chip. Results We established such models with 15 nude mice. Tumor taking rate was 100% ( 15/15 ) on the wall of gastric. The metastasize to intrapcritoneal cavity were 40% (6/15) . Analysed the results by the gene chip and calculated the Ratio (Cy3/Cy5). The gastric cancer cell and gastric cancer metastasize cell were differentially expressed as follows: up 192 genes, down 139 genes. Conclusions We used the gene chip technology to select the hundreds of peritoneal metastasis of gastric cancer of the differentially expressed genes, The experimental provide a basis and reference for study the genes about the metastasize of gastric cancer.
