基质金属蛋白酶-1基因多态性与结直肠癌易感性的荟萃分析

摘　　要：目的探讨通过系统评价人群MMP-1基因多态性在结直肠癌发生中的作用以揭示两者之间的相关性。方法通过检索MEDLINE、Embase、Cancerlit、AACR(American Associationfor Cancer Research)、中国生物医学文献数据库(CBMdisc)、中国知网、万方数据库等数据库,得到了5个关于MMP-1基因多态性和结直肠癌相关性的病例对照研究;利用Q-检验和I2检验以检验异质性,利用随机效应模型和固定效应模型以计算总的比值比(OR)、95%可信区间(CI)和总的效应P值。结果共选共5个病例-对照试验(case-control study)研究,其中包含599个结直肠癌病例和829个正常对照,MMP-1 1607 2G2G基因型和合并后的*/2G基因型与结直肠癌发生相关性的OR和CI分别是[OR1.24,95%CI(0.79,1.96);P=0.35]和[OR1.66,95%CI(1.14,2.42);P=0.008]。结论 MMP-1 1607 2G2G基因型与结直肠癌发生可能无相关性,而2G等位基因携带者与结直肠癌的易感性显著相关。Objective To investigate the role in the occurrence of colorectal cancer through systematic evaluated population matrix metalloproteinase-1 gene polymorphisms and to examine the correlation between MMP-1 genotype and colorectal cancer.Methods Searching database such as the MEDLINE,Embase,Cancerlit,AACR（American Association for Cancer Research）,CBMdisc,CNKI,VIP,WANFANG DATA etc,yielded five case-control studies that reported on the correlation between MMP-1 polymorphisms and colorectal cancer.Using Q-test and I2-test,the heterogeneity was detected and using random effect modle and fixed effect modle,the total odds ratios（OR）,95% confidence intervals（CI）and total effect P value were calculated.Results Total of 5 cases-control study was enrolled,including 599 cases with colorectal cancer and 829 cases served as normal control,which MMP-1 1607 2G2G genetype post-combined ＊/2G genetype with OR and CI of occurrence correlation of colorectal cancer were OR1.24,95% CI0.79,1.96 P =0.35 and 1.66,95% CI：1.14,2.42 P=0.008 respectively.Conclusion MMP-1 1607 2G2G genetype may be not correlated with the occurrence of colorectal cancer,but the 2G allele carriers are significantly correlated with the susceptibitity of colorectal cancer.
