摘  要:目的探讨GADD45β和survivin基因在人原发性肝癌(HCC)组织中的蛋白表达及意义。方法收集不同分化程度的HCC组织标本68例,正常肝组织标本20例,采用免疫组化S-P法检测GADD45β和survivin基因的蛋白表达。结果 GADD45β基因与survivin基因在HCC组织中的蛋白表达率分别为38.24%(26/68)和75.00%(51/68),两者的表达均与HCC的病理分化程度、临床分期有关(P<0.05),survivin基因的表达还与门脉癌栓有关(P<0.05)。随着survivin基因在HCC组织中蛋白表达率的升高,GADD45β基因蛋白表达强度明显下降,两者的表达呈负相关(r=-0.454,P<0.05)。结论 GADD45β基因在HCC发生发展中起抑制作用,GADD45β基因表达缺失是HCC发生发展的重要因素之一;survivin基因的异常表达在肝癌的发生发展中起促进作用。联合检测GADD45β基因和survivin基因的蛋白表达有助于判断HCC的恶性程度和HCC患者的预后。Objective To study the expressions and clinical significances of GADD45β and survivin gene in human hepatocellular carcinoma(HCC).Methods Immunohistochemical method was used to detect the expressions of GADD45β and survivin in both HCC and normal liver tissues(68 cases of HCC and 20 cases of normal liver tissues).Results The positive rates of GADD45β and survivin expressions in HCC tissues were 38.24%(26/68) and 75.00%(51/68) respectively.The expressions of GADD45β and survivin were both closely correlated with the differentiation degree and clinical stage of HCC(P0.05).The expressions of survivin in HCC tissues were also related to tumor thrombus of portal vein.And the expressions of GADD45β had negative correlation with the expressions of survivin in HCC tissues(r=-0.454,P0.05).Conclusion GADD45β may inhibit the occurrence and development of HCC,but survivin may have the opposite effect.Combined detection of GADD45β and survivin expressions may be beneficial to determine the malignancy degree and predict prognosis of HCC.
