摘 要:目的 间接了解Alzheimer’s 病(AD)海马神经元总体基因表达水平。方法 10 例AD 患者病理脑海马组织经Nissl’s 染色后,图像分析神经元核仁、核浆比值,核仁组成区银染色后图像分析神经元着色面积及积分吸光度,并与相应的正常青年组、老年组各10 例的海马标本进行比较对照。结果 老年组和AD组海马神经元核浆比值( 分别为541 ±81 及528 ±97) 下降与青年组(608 ±59) 相比,差异有显著性( P< 005 及001) ;老年组与AD组相比,差异无显著性( P> 005) 。同时,老年组和AD组海马神经元核仁着色面积减少,积分吸光度减弱;老年组和AD组相比,AD组海马神经元核仁着色面积(2117 ±548um) 及积分吸光度(17324 ±4281) 相对增强,与老年组( 分别为1806 ±661um 与14143 ±5499) 相比,差异有显著性( 均为P< 005) 。结论 Nissl’s 染色说明老年人和AD 患者海马神经元功能低下,核仁组成区银染色提示老年人和AD 患者神经元rDNA转录活性低下,但AD患者海马神经元转录活性相对增高,其细胞总体基因表达活性相对增强,此种细?Objective To determine indirectly the degree of total gene expression in the hippocampal neurons of Alzheimer's disease (AD).Methods Neuropathological examination was carried out in 10 cases of AD and control groups,including the Yang and elderly patients without any neurological disorder.The nucleoli nucleoplasm ration,stained area and intergral optical density of silver stained nucleolar orgnizer regions (NORS) in the hippocampal neurons studied by image analysis after Nissl's and silver stainings.Results The nucleoli nucleoplasm rations of hippocampal neurons were decreased in both of AD and elderly groups,in comparision with young group,with significant difference statistically ( P <0.05 and P <0.01 respectively).There was no significant difference between the AD and elderly group ( P >0.05) The stained area and intergral optical density in hippocampal neurons were also reduced in the AD and elderly groups as compared with young group.But the stained area and intergral optical density of hippocampal neurons were increased more significantly in the AD group than that in the elderly group ( P <0.05).Conclusions The results of nucleoli nucleoplasm rations indicated that the hippocampal neuron functions in the AD and elder were declined because of degenerative process of neurons.The results of silver stained NoRs suggested that more active translating activity of rDNA in the AD hippocampal neurons might represent the cell defence function although the translating activity was diminished in the AD and elderly groups.
