鼻咽癌患者机体TCR Vβ亚家族取用格局与预后相关性研究

摘　　要：目的研究鼻咽癌(NPC)患者机体TCR Vβ24个亚家族的表达格局情况,探讨其与肿瘤分期及预后的关系。方法采用流式细胞仪技术,检测44例NPC各期放疗前患者机体TCR Vβ24个亚家族的表达情况,并与18例正常对照组人群进行对比,分析TCR Vβ24个亚家族各指标的表达情况与肿瘤分期及预后的关系。结果 (1)NPC组同正常对照组相比,TCR Vβ1、Vβ7.1、Vβ7.2、Vβ9、Vβ13.1等表达明显升高(P<0.05),而TCR Vβ5.2、Vβ5.3和Vβ23表达则显著降低(P<0.05);(2)TCR Vβ24个亚家族中TCR Vβ5.2、Vβ5.3、Vβ9、Vβ11、Vβ13.1、Vβ13.2及Vβ18等差异性表达(P<0.05)与NPC肿瘤TNM分期及临床分期有密切关系;(3)5年生存患者与5年死亡患者相比体内TCR Vβ5.2、Vβ13.2及Vβ18的表达差异具有统计学意义(P<0.05),而5年无瘤生存患者与5年复发转移及死亡患者相比,体内TCR Vβ5.2、Vβ9、Vβ11、Vβ13.2及Vβ18表达差异具有统计学意义(P<0.05)。结论 TCR Vβ24个亚家族取用格局差异与NPC的TNM分期及肿瘤临床分期存在相关性,并可能作为NPC肿瘤患者判断预后的前瞻性指标。Objective To analyze the expression of TCR Vβ in different clinical stages of NPC;and the relation with prognosis.Methods Forty-four specimens of pathologically confirmed different clinical stages NPC were tested,18 normal tissue as comparison group.The expression of TCR Vβ were examined by flow cytometry （FCM）,and then analyzed the relationships between two factors with the patients＇ prognosis and with patients＇ clinical pathologic features.Results The expressions of TCR Vβ1,Vβ7.1,Vβ7.2,Vβ9,Vβ13.1 in NPC were higher than the normal tissue （P0.05）,but TCR Vβ5.2,Vβ5.3 and Vβ23 were lower than the normal tissue;The expression of TCR Vβ5.2,Vβ5.3,Vβ9,Vβ11,Vβ13.1,Vβ13.2 and Vβ18 were correlated with the TNM stages of NPC（P0.05）;The differences of TCR Vβ5.2,Vβ13.2 and Vβ18 expression between 5-year survival group and 5-year death group were great significant （P0.05）.And the differences of TCR Vβ5.2,Vβ9,Vβ11,Vβ13.2 and Vβ18 expression between 5-year survival without tumor group and non-5-year survival without tumor group were great significant （P0.05）.Conclusions The expressions of TCR Vβ family in NPC have relationship with the TNM stages and clinical stages,can be used as prognostic molecular markers in NPC patients.
