摘  要:目的观察抑肽酶对凝血酶所致大鼠星形胶质细胞损伤的保护作用。方法将体外培养的大鼠星形胶质细胞随机分为正常对照组、凝血酶组及抑肽酶干预组。24h后,测定培养液中乳酸脱氢酶(LDH)活性。四甲基偶氮唑盐(MTT)比色分析法比较细胞损伤程度(OD值)。用末端脱氧核糖核苷酸转移介导的dUTP缺口末端标记(TUNEL)检测不同抑肽酶浓度作用培养星形胶质细胞后的凋亡细胞。结果抑肽酶干预组的OD值(0.67±0.11,0.78±0.07和0.80±0.13)高于凝血酶组的OD值(1.05±0.09)(P<0.05);抑肽酶干预组的LDH活力(154.31±10.74、101.38±13.59和92.72±11.92)明显低于凝血酶组的LDH活力(267.09±25.35)(P<0.05)。抑肽酶干预组的星形胶质细胞凋亡数(13.20±1.52、9.10±1.20和4.80±0.89)明显低于凝血酶组星形胶质细胞凋亡数(26.10±1.82)(P<0.05)。结论抑肽酶可能是通过减轻凝血酶诱导星形胶质细胞损伤并增加其活力、减轻其凋亡,对其实施保护作用。Objective To observe the protective effect of aprotinin on astrocytes damaged by thrombin.Methods Cultured astrocytes of rats were divided into five groups randomly,normal control group,thrombin group and aprotinin pretreatment group.After twenty-four hours,the activity of lactate dehydrogenase was determined by chemistry reaction in the culture system.The viability of astrocytes(OD value)was measured by methylthiazolyl tetrazolium(MTT).The apoptotic cells in different aprotinin concentrations were detecte by TUNEL.Results the OD values in the intervention groups of aprotinin(0.67±0.11,0.78±0.07 and 0.80±0.13)were higher than the thrombin group(1.05±0.09)(P0.05);The LDH activities in the intervention groups of aprotinin(154.31±10.74,101.38±13.59 and 92.72±11.92)were significantly lower than the thrombin group(267.09±25.35)(P0.05).The apoptotic cells in the intervention groups of aprotinin(13.20±1.52,9.10±1.20 and 4.80±0.89)was significantly lower than the thrombin group(26.10±1.82)(P0.05).Conclusions Aprotinin can alleviate the astrocytes damage induced by thrombin by increasing viability and decrease apoptosis of astrocytes,to achieve its protective effect.
