摘  要:目的观察大鼠单侧输尿管完全梗阻(complete unilateral ureteral obstruction,CUUO)后不同时期肝细胞生长因子(hepatocyte growth factor,HGF)表达的变化,探讨HGF在肾脏间质纤维化发病机制中的意义。方法建立CUUO模型,采用半定量逆转录聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)检测和免疫组化法检测(1周、2周、4周)梗阻侧和代偿侧肾脏及假手术对照组HGFmRNA和HGF蛋白的表达。结果 CUUO模型梗阻侧和代偿侧肾脏HGFmRNA及HGF蛋白的表达在输尿管结扎后1周、2周、4周不同时间点均显著高于对照组肾脏(P<0.05)。随着梗阻时间的延长,HGFmRNA(P<0.05)和HGF蛋白(P<0.05)逐渐增加。而且对于同一个实验对象代偿侧要远远高于梗阻侧HGFmRNA(P<0.05)和HGF蛋白(P<0.05)。结论在整体条件下随着肾脏间质纤维化程度的加重,HGF在有功能的残余肾脏组织中表达逐渐增强,提示HGF可能在肾脏纤维化的发病机制中起重要的作用。Objective To investigate the role of HGF activation pathway in the development of renal tubule interstitial fibrosis in vivo.Methods Rats were sacrificed after1 weeks,2 weeks,and 4 weeks of complete unilateral ureteral obstruction(CUUO),and sham-operated rats were sacrificed as the control.HGFmRNA and protein and were analyzed with semi-quantitative reverse transcription polymerase chain reaction(RT-PCR) and Immunohistochemistry.Results HGFmRNA and protein levels of the CUUO group at the 1 weeks,2 weeks,4 weeks in the kidneys of rats with obstructed kidneys and compensatory kidneys,were significantly increased(P0.05),compared to sham-operated rats.HGFmRNA expression were peaked 7 days after CUUO(P0.05).HGF protein expression was peaked 1 week after CUUO.The levels of HGF protein expression were peaked 4 weeks after CUUO(P0.05).Conclusion Expressions of HGF are increasted in renal tissue after CUUO.HGF signaling pathway may be involved in CUUO rats and plays a concordant role in the pathogenesis of renal interstitial fibrosis.
