摘  要:目的:研究牛膝对P388白血病细胞的体外细胞毒性及其活性成分。方法:牛膝根以水提取,经Toyopear1HW-40C凝胶过滤,得到高分子物质部分。以P388白血病细胞的体外培养确定细胞毒活性。以SephacrylS-200进行分离和纯化。以1H-NMR进行初步的组成鉴定。结果:牛膝水提取物的高分子物质部分有细胞毒性,IC50值为5.2μg·ml-1。多糖为其主要成分,由木糖、甘露糖和果糖等单糖做成。结论:牛膝水提取物的高分子物质部分对P388白血病细胞有显著的体外细胞毒性。多糖可能是细胞毒性成分。OBJECTIVE:To study the active composition of the extract from Achyranthes bidentata root.Bl.(amaranthaceae) and its cytotoxicity against P388 leukemia cells in vitro. METHODS:Water macerating extract of A.bidentata root was obtained and isolated by Toyopearl HW 40C gel filtration to get a high molecular part. In vitro cytotoxicity was based on culturing P388 leukemia cells.Active composition was seperated and purified with Sephacryl S-200 from the high molecular part and identified with 1 H NMR spectrum.RESULTS:Cytotoxicity (IC 50 5.2μg·ml -1 ) was detected and a polysaccharide was found to be a main composition of the high molecular part,which was composed of xylose,mannose,fructose and so on.CONCLUSION:High molecular part from water macerating extract of A.bidentata root has a remarkable cytotoxicity against P388 leukemia cells in vitro. The polysaccharide of A.bidentata root may be cytotoxic.
