左旋棉酚体外诱导人前列腺PC-3细胞凋亡的研究

摘　　要：目的:研究左旋棉酚对人前列腺癌PC-3细胞体外增殖和凋亡的影响。方法:MTT法检测左旋棉酚对PC-3细胞增殖的影响,透射电镜观察细胞超微结构的改变,TUNEL染色观察细胞凋亡,流式细胞术检测细胞周期的改变,半定量RT-PCR检测Bcl-2及Bak mRNA的表达水平。结果:左旋棉酚能显著抑制PC-3细胞的体外增殖,并呈浓度-时间依赖性。左旋棉酚可以诱导PC-3细胞发生典型的凋亡形态学改变和G0/G1期阻滞,RT-PCR显示Bcl-2 mRNA表达水平降低,Bak mRNA表达水平升高。结论:左旋棉酚能诱导前列腺癌细胞的体外凋亡,主要可能与Bak mRNA的表达升高及Bcl-2 mRNA的表达下调有关。Objective：To investigate the effect of （ - ）gossypol on the proliferation and apoptosis in human prostate cancer PC - 3 cells in vitro. Methods： The cell proliferation was evaluated by methyl thiazolyl tetrazolium （M3T） assay. Cell ultramicro - structure was studied by transmission electron microscope. The morphology of apoptotic cells was observed by TUNEL staining , cell cycle and apoptosis were detected by flow cytometry （ FCM ）. The Bcl - 2 and Bak mRNA level were measured by semiquantitative RT - PCR. Rusults ： （ - ） Gossypol significantly inhibited the proliferation of PC - 3 cells in a dose - and time - dependent manner. The PC - 3 ceils were arrested in G0/G1 phase and showed typical apoptosis characteristics in FCM and transmission electron microscope analysis. （ - ） Gossypol down - regulated Bcl - 2 mRNA and up - regulated Bak mRNA level. Conclusion ： （ - ） Gossypol can induce apoptosis of human prostate cancer PC - 3 cells in vitro and the main reason may be related with the up - regulation of Bak mRNA and the down - regulation of Bcl - 2 mRNA.
