摘　　要：目的从基因转录水平确定人视网膜色素上皮（ＲＰＥ）细胞是否具有合成纤溶酶原激活物抑制１（ＰＡＩ１）蛋白酶的生物学特性。方法取供体人眼ＲＰＥ细胞进行体外培养；当细胞生长融合后抽取ＲＮＡ，应用Ｎｏｔｈｅｒｎｂｌｏｔ分析技术，经特异的探针对人ＲＰＥ细胞的总ＲＮＡ进行检测。结果在体外培养的人ＲＰＥ细胞基因转录产物中存在有ＰＡＩ１ｍＲＮＡ的表达，其长度分别为３．０ｋｂ和２．３ｋｂ。结论人ＲＰＥ细胞能够合成ＰＡＩ１蛋白酶；它可能在一些致盲性眼病的病理发生过程中起到重要的作用。ObjectiveTodeterminewhetherhumanretinalpigmentepithelial(RPE)cellscanphysiologicallysynthesizeplasminogenactivatorinhibitor1(PAI1)atthetranscriptionlevel.MethodsHumanRPEcellsfromdonoreyewereisolatedandculturedinvitro.TotalRNAwasisolatedwhenRPEcellsinprimaryculturebecameconfluent.PAI1messengerRNA(mRNA)wasidentifiedbyNothernblotanalysisusingspecificcDNAprobes.ResultsTherewereexistenceforexpressionofPAI1mRNAinthegenetranscriptionproductsfromhumanRPEcells.ThesizesofmRNAwere3.0kband2.3kbforHumanRPEcellstranscripts.ConclusionHumanRPEcellscansynthesizePAI1enzym,whichmayplayanimportantroleinthedevelopmentofseveralblindingoculardisorders.
