摘 要:目的探讨窖蛋白1(Caveolin-1)蛋白及mRNA在食管鳞状细胞癌(ESCC)组织中的表达及其意义。方法应用免疫组织化学及原位杂交方法检测57例ESCC组织、34例癌旁不典型增生组织及57例正常食管黏膜组织中Caveolin-1蛋白及mRNA的表达。结果 ESCC组织、癌旁不典型增生组织及正常黏膜组织中Caveolin-1蛋白的阳性表达率依次降低,分别为54.4%(31/57)、38.2%(13/34)和19.3%(11/57),组间比较差异均有统计学意义(P<0.05);ESCC组织、癌旁不典型增生组织及正常黏膜组织中Caveolin-1 mRNA的阳性表达率依次降低,分别为64.9%(37/57)、47.1%(16/34)和22.8%(13/57),组间比较差异均有统计学意义(P<0.05);Caveolin-1蛋白及mRNA的表达均与ESCC的分化程度、浸润深度和淋巴结转移密切相关(P<0.05)。结论 Caveolin-1蛋白及mRNA表达与ESCC的发生、发展和转移密切相关,可作为ESCC的一种标记物,联合检测Caveolin-1蛋白及mRNA在评估ESCC预后方面具有一定的临床价值。Objective To investigate the expressions of Caveolin-1 protein and mRNA in esophageal squamous cell carcinoma(ESCC)tissues and its significance.Methods The expressions of Caveolin-1 protein and mRNA was detected by immunohistoche-mistry and in situ hybridization methods in 57 cases of ESCC tissues,34 cases of adjacent atypical hyperplasia epithelial tissues and 57 cases of normal esophageal epithelial tissues.Results The positive expression rate of Caveolin-1 protein gradually decreased from 54.4%(31/57)in ESCC,38.2%(13/34)in adjacent atypical hyperplasia epithelium to 19.3%(11/57)normal esophageal epithelium specimens,and there was a significant statistic difference among the groups(P0.05).Additionally,the expression rates of Caveolin-1 mRNA decreased by turns from ESCC,adjacent atypical hyperplasia epithelium to normal esophageal epithelium specimens,which were 64.9%(37/57),47.1%(16/34)and 22.8%(13/57)respectively,and there was a significant statistic difference among the groups(P0.05).The expressions of Caveolin-1 protein and mRNA were closely correlated with the different differentiation levels,infiltrative depth and lymph node metastasis in ESCC tissues(P0.05).Conclusion The expressions of Caveolin-1 protein and mRNA may play important roles in the infiltration,metastasis and carcinogenesis of ESCC,suggesting it may be used as a tumor maker of ESCC.Combined detections of Caveolin-1 protein and mRNA have some clinical value in assessing the prognosis of ESCC.
