新生大鼠大脑皮层神经元原代培养方法

摘　　要：目的:介绍一种简单、生长良好的大脑皮层神经元的培养方法。方法:急性分离新生鼠大脑皮层制作单细胞悬液进行原代培养,观察培养神经元的生长形态,并用免疫荧光化学法对其进行鉴定。结果:培养的神经元细胞清晰,光晕明显,生长良好,免疫荧光鉴定结果表明该方法培养的细胞纯度较高,能够达到实验研究的要求。结论:该培养方法简单、可靠,是神经元培养的良好方法。Objective：To introduce a simple and good method of culturing cortical neurons.Methods：The cerebral cortexes of newborn rats were separated acutely and made into singl cell suspension for primary culture.The shapes of cultured neurons were observed during growth process.Immunofluorescence method was used to identify them.Results：The cultured neurons were vivid.The cultured neurons grew well and the shapes were clear.Immunofluorescence results show that the cultured cells with high purity can be used for experimental research.Conclusion：The culturing method is simple,reliable and good.
