摘 要:目的研究异体脱矿骨-胶原蛋白-四环素复合材料促进兔下颌骨牵张成骨作用。方法将四环素和胶原蛋白包被于兔脱矿骨颗粒上,制备成兔脱矿骨-胶原蛋白-四环素复合材料。选用36只新西兰大白兔,随机分为三组,每组12只,A组为对照组,B组为脱矿骨组,C组为脱矿骨-胶原蛋白-四环素复合材料组。建立单侧下颌骨牵张模型,术中即牵张2 mm间隙。A组间隙注入0.5 ml生理盐水,B组和C组分别在间隙中填充0.5g的脱矿骨颗粒和脱矿骨-胶原蛋白-四环素复合材料。在牵张后固定期第1、3、6周分别进行X线、组织学检查及血清碱性磷酸酶活性及骨钙素含量、骨密度、生物力学检测。不同组间均值比较采用t检验,SPSS 13.0统计软件包统计分析。结果 C组的新骨成骨速度和质量明显好于A、B两组,B组的新骨成骨速度和质量好于A组;各检测指标,除固定期第3、6周,C组和B组血清骨钙素含量没有显著性差异外,三组间均有显著性差异(p<0.05)。结论异体脱矿骨-胶原蛋白-四环素复合材料能够明显促进兔下颌骨牵张成骨。Objective To investigate the accelerative effect of allogeneic demineralized bone -collagen - tetracycline composite material on the mandibular distraction osteogenesis (DO) in rabbits. Methods Tetracycline and collagen were coated on particles of rabbit demineralized bone to prepare rabbit demineralized bone - collagen - tetracycline composite material. Thirty six New Zealand white rabbits were randomly divided into three groups, n = 12, group A as the control group, group B as the demineralized bone group and group C as the demineralized bone - colla- gen- tetracycline composite group. The unilateral mandibular distraction model was estabilished in rabbits, and the distraction gap was stretched to 2 mm during the surgery. 0. 5 ml phyological saline was injected into the gap in group A rabbits ,0. 5 g demineralized bone particles and demineralized bone -collagen -tetracycline composite material were seperately filled into the gaps in group B and group C rabbits. The rabbits were sacrificed at the end of 1,3,6 weeks after the completion of distrction for X - ray, histological examinations and serum alkaline phosphatase activity and os- teoealcin content,bone mineral density,biomechanical testing respectively. SPSS 13.0 software package and Student' s t test were used for statistical analysis. Results [n the rabbits of group C, the formation rate and the quality of new generated bone were superior to those of the other two groups. The formation rate and the quality of new generated bone of group B rabbits were superior to those of the group C rabbits. The detection indexes of the three groups were signifi- cantly different (p 〈 0. 05 ) exception for the serum ostecocalin contents of the gourp B and group C rabbits at the end of 3 and 6 weeks during fixation. Conclusion The allogeneic demineralized bone - collagen - tetracycline composite materials can significantly promote the rabbit mandibular distraction osteogenesis.
