摘 要:初步探讨了大孔树脂纯化碱提花生壳总黄酮的工艺条件,对大孔树脂的种类及其静态吸附、解吸附条件进行初步探讨。通过静态吸附和解吸附的比较,从6种不同型号的大孔吸附树脂中选出DM301进行静态吸附解吸动力学,发现其吸附解吸平衡时间分别为3 h和5 h。通过单因素实验,DM301的最佳吸附条件为20℃、pH8.5,样液中花生壳总黄酮初始浓度为(0.138±0.01)mg/mL;最佳解吸条件为解吸液乙醇浓度80%,解吸液pH9.5,解吸液用量7.5 mL/g湿树脂。This article studied the purificating conditions of total flavonoids from peanut hull with macroporous resin.Preliminary discussion and research was made on types of macroporous resin and its adsorption and desorption conditions.Through the comparison of static adsorption and desorption,macroreticular resins DM301 was selected from six different types of macroreticular resins to carry out static adsorption and desorption kinetics,it was found that the equilibrium time of adsorption and desorption were separately 3 h and 5 h.Through the single factor experiment.The best adsorption conditions of DM301 were determined as follows: the temperature of adsorption is 20 ℃,the pH value is 8.5,the initial concentration of total flavonoids from peanut hull in sample solution is(0.138±0.01) mg/mL.The best desorption conditions were obtained as follows: the concentration of ethanol desorption liquid is 80%,the pH value is 8.5,the volume of desorption liquid is 7.5 mL/g wet resin.
