摘  要:目的:寻找山茱萸中具有抑制α-葡萄糖苷酶活性的成分。方法:用有机溶剂进行提取,在体外建立微量酶反应体系,以4-硝基苯-α-D-吡喃葡萄糖苷PNPG作为底物,阿卡波糖为阳性对照,检测山茱萸不同溶剂萃取物对α-葡萄糖苷酶抑制活性。结果:山茱萸石油醚萃取物[50%抑制浓度(IC50)=38.90 mg·L-1]、乙酸乙酯萃取物(IC50=2.40 mg·L-1)和正丁醇萃取物(IC50=4.26 mg·L-1),远低于阳性对照阿卡波糖(IC50=1 081.27 mg·L-1)。结论:山茱萸各萃取物均具有很好的α-葡萄糖苷酶抑制活性,且乙酸乙酯萃取物的抑制活性最好。Objective:To search α-glucosidase inhibitors from Cornus officinalis. Method: All the extracts were extracted by organic solvent and the trace enzyme reaction system was constructed in vitro. Taking PNPG as substrate,acarbose as positive control,and inhibitive effect of extracts from C. officinalis on α-glucosidase was detected. Result: The IC50 value of petroleum ether extracts ( 38.90 mg.L - 1 ), ethyl acetate extracts ( 2.40 mg. L- 1 ) and n-butanol extracts(4.26 mg.L-1) were far lower than that of acarbose( 1 081.27 mg.L-1) as positive control. Conclusion:All the extracts of C. officinalis showed a better α-glucosidase inhibitory activity and ethyl acetate extracts showed the strongest inhibitory activity on α-glucosidase.
