摘  要:通过山葵愈伤组织的诱导培养,测定培养物中的风味物质,为通过组织培养的手段来生产药用成分提供依据。选择山葵幼叶和叶柄作为外植体,以MS为基本培养基,配以不同浓度的激素进行愈伤组织的诱导。选择继代几次以后的愈伤组织进行风味物质的测定。确定MS+6-BA 1.0+NAA 3.0为愈伤组织诱导的最佳条件,致密型的愈伤组织经过细胞悬浮培养以后收获测定其异硫氰酸酯类含量明显高于松散型的愈伤组织。用外植体诱导出山葵愈伤组织,确定了最佳诱导条件,并且对于细胞悬浮收获物的异硫氰酸酯类含量的测定也得到了最佳值。The callus of Wasabia was cultured in induction medium,and the flavor components were measured in the culture.It will provide the basis for the production of pharmaceutical components.The explants,the spire and petioles were cultured in the MS with different concentrations of hormones for callus induction.The flavors of culture were measured after subculture callus.The best recipe of the culture medium was: MS+6-BA1.0mg/mL+NAA3.0mg/mL.Isothiocyanates' content was significantly higher by cell suspension culture in compact type of callus than that in loose callus.The results showed that the best recipe of the culture medium was concluded by inducing the explants,and the best value of isothiocyanates content in the cell suspension has also been determined.
