摘  要:目的检测兔干眼动物模型角膜组织中基质金属蛋白酶-1(matrix metalloprotein-ase,MMP-1)和MMP-9的表达,探讨MMP在干眼发病机制中的作用。方法新西兰大白兔共30只,采用摘除泪腺、第3眼睑及Harders'腺的方法造模。造模前及造模后第3天、第5天、第7天、第14天、第21天、第28天行ShirmerⅠ试验和荧光素角膜染色检查。分别于造模后第14天、第28天各取15只实验兔角膜组织行免疫组织化学检查。结果造模前兔ShirmerⅠ试验值(16.4±3.8)mm,造模后第3天、第5天、第7天、第14天、第21天、第28天分别为(13.1±3.2)mm、(10.5±2.2)mm、(8.4±1.7)mm、(6.5±2.1)mm、(5.6±2.4)mm、(5.2±2.3)mm,造模后ShirmerⅠ试验值随着时间的推移逐步下降,7d后稳定在10mm以下。造模后各时间点ShirmerⅠ试验值与造模前比较均存在显著性差异(均为P<0.01)。造模前兔角膜荧光素染色阴性,造模后出现阳性染色,并随着时间的推移染色程度逐渐加重。造模后第14天、第28天术眼角膜光学显微镜下可见明显组织结构异常,且MMP-1和MMP-9的表达呈高度相关性,随时间的推移均显著提高。结论 MMP-1和MMP-9均参与了干眼的病理发展过程。Objective To investigate the expression of collagenase matrix metalloproteinase(MMP-1)and gelatinase MMP-9 in the rabbit corneal tissues of dry eye model at different periods and to elucidate the functions of MMP in the pathological progresses of dry eye.Methods The dry eye models were established on the right eyes of 30 rabbits,after ablating the corresponding lacrimal gland,the tertiary eyelid and the Harders' gland.The Schirmer I test and corneal fluorescein staining were used to evaluate the inner situations of the eyes at different time points at the 3rd,5th,7th,14th,21st and 28th day after modeling.The corneal tissues of 15 rabbits were analyzed with immunohistochemistry at the 14th and 28th day after modeling,respectively.Results The mean Shirmer Ⅰ test value was(16.4±3.8)mm before modeling,were(13.1±3.2)mm,(10.5±2.2)mm,(8.4±1.7)mm,(6.5±2.1)mm,(5.6±2.4)mm and(5.2±2.3)mm at the 3rd,5th,7th,14th,21st and 28th day after modeling.The Shirmer l test value kept decreasing after modeling and became stable within 10 mm at the 7th day after modeling.There was no significant difference in Shrimer Ⅰ test value for each time point before modeling comparing with that before modeling(all P0.01).Corneal fluorescein staining was negative before modeling and positive after modeling.The degree of staining increased with the time.Obvious structural abnormal was found under corneal light microscope at the 14th day and 28th day after modeling.The expression of MMP-1 and MMP-9 was highly correlated,improved with time and positively correlated with severity of dry eye.Conclusion MMP-1 and MMP-9 are involved in the pathological progresses of dry eye.
