摘 要:目的:探讨Myocardin在维甲酸(RA)诱导骨髓间充质干细胞(BMSC)向神经细胞(NC)分化过程中的作用及与维甲酸X受体α(RXRa)的相互作用。方法:免疫细胞荧光检测RA诱导BMSC向NC分化;RT-PCR检测My-ocardin及RXRa在RA诱导BMSC分化为神经细胞过程中的表达;萤光素酶活性实验分析Myocardin和RXRa对EGR-4-luc的影响。结果:RA可有效诱导BMSC向NC分化;在mRNA水平上,RA能显著增加RXRa、Myocardin的表达,且在一定范围内呈剂量依赖;Myocardin与RXRa协同促进EGR-4-luc的表达。结论:Myocardin在BMSC向NC分化过程中起重要的正向调控作用,这一作用是与RXRa协同完成的。Objective: To discover the role of myocardin in the differentiation of bone marrow stromal cells(BMSC) into neural cells under induction by retinoic acid(RA), and investigate the cooperation with retinoid X receptor alpha(RXRa). Methods: Expression of NSE and MAP-2 in BMSC treated with RA were tested by immunohistochemcal assay; mRNA expression level of myocardin and RXRa in differentiation of BMSC into neural cells were tested with RT-PCR. The luciferase assay was used to investigated the effection of RXRa and myocardin to EGR-4-luc. Results: BMSC could be effectively induced into neural ceils by RA, and the mRNA expression level of myocardin and RXRa were upregulated. The cooperation of RXRa and myocardin could regulate the expression level of EGR- 4-luc. Conclusion: Myocardin may play an important role in the differentiation of BMSC into neural cells. This function may be related to the cooperation between RXRa and myocardin.
